探讨草苁蓉环烯醚萜(IGBR)对二乙基亚硝胺(DEN)诱发的肝癌前病变大鼠血清标志酶及抗氧化活力的影响。按改良的Solt-Faber氏方法复制大鼠肝癌前病变模型,用IGBR饲养大鼠6周后,ELISA法测定血清肿瘤坏死因子-α(TNF-α),比色法测定γ-谷酰胺转肽酶(γ-GT)、谷胱甘肽-S-转移酶(GST)、丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)活性及丙二醛(MDA)和一氧化氮(NO)含量。结果表明,IGBR降低肝癌前病变大鼠血清γ-GT、GST、ALT、AST活性,升高血清SOD、CAT、GSH-PX活性和降低MDA水平。同时,IGBR增高血清NO水平,但对血清TNF-α水平没有显著影响。提示,IGBR具有明显的抗致癌作用,其作用机制可能是通过增高抗氧化活性来实现的。IGBR对肿瘤免疫的影响需进一步探讨。
To investigate the effect of IGBR on serum enzyme and anti-oxidative activity of rats with hepatocarcinoma induced by diethylnitrosamine (DEN). Rat model of precancerous lesions was reproduced by modified Solt-Faber’s method. Rats were fed with IGBR for 6 weeks. Serum tumor necrosis factor-α (TNF-α) was measured by ELISA and γ-glutamyl transpeptidase Glutathione-S-transferase (GST), alanine aminotransferase (ALT), aspartate aminotransferase (AST), superoxide dismutase (SOD) Catalase (CAT), glutathione peroxidase (GSH-PX) activity and malondialdehyde (MDA) and nitric oxide (NO) content. The results showed that IGBR decreased the serum levels of γ-GT, GST, ALT and AST, increased the activities of serum SOD, CAT, GSH-PX and decreased the level of MDA in rats with precancerous lesions. Meanwhile, IGBR increased serum NO levels but had no significant effect on serum TNF-α levels. Tip, IGBR has a significant anti-carcinogenic effect, its mechanism may be achieved by increasing antioxidant activity. The effect of IGBR on tumor immunity needs to be further explored.
