目的:研究核糖核苷酸还原酶M2(RRM2)在结肠直肠癌组织的表达及对癌细胞增殖和迁徙的作用。方法:应用组织芯片免疫组化技术测组织中RRM2的表达。用siRNA转染RRM2高表达的细胞株HCT116以验证干扰效果,CCK-8法检测细胞增殖能力,Transwell实验评估转染后癌细胞迁移能力。结果:结肠直肠癌组织中RRM2的表达明显高于癌旁正常组织(P<0.05),且与浸润深度、分化程度、TNM分期均有相关性;在结直肠细胞株中,HCT116细胞RRM2在mRNA和蛋白水平表达量最高,用干扰效率最高的siRNA下调RRM2表达可以明显抑制HCT116细胞的增殖能力。在Transwell迁徙实验中,RRM2干扰组穿膜细胞数为(11±2)个,显著低于阴性对照组[(29±4)个]和空白对照组[(23±3)个](P<0.01)。结论:数据显示RRM2在结肠直肠癌组织中呈过表达,且在结肠直肠癌进展和预后发挥重要作用,其有望成为评估结肠直肠癌进展及预后的潜在指标。
Objective: To study the expression of ribonucleotide reductase M2 (RRM2) in colorectal cancer and its effect on the proliferation and migration of cancer cells. Methods: Tissue chip immunohistochemistry was used to detect the expression of RRM2. The high expression of RRM2 was transfected into HCT116 cells to verify the interference effect. The cell proliferation was detected by CCK-8 assay and the transwell assay was used to evaluate the ability of cancer cells to transfect. Results: The expression of RRM2 in colorectal cancer tissues was significantly higher than that in adjacent normal tissues (P <0.05), and was correlated with the depth of invasion, differentiation degree and TNM stage. In colorectal cancer cell lines, the expression of RRM2 in mRNA of HCT116 And protein levels were the highest. Down-regulation of RRM2 expression by siRNA with the highest interference efficiency could significantly inhibit the proliferation of HCT116 cells. In transwell migration experiments, the numbers of transmembrane cells in the RRM2 interference group were (11 ± 2), significantly lower than those in the negative control group (29 ± 4) and the blank control group (23 ± 3) (P < 0.01). CONCLUSIONS: The data show that RRM2 is overexpressed in colorectal cancer tissues and plays an important role in the progression and prognosis of colorectal cancer, which is expected to be a potential indicator for assessing the progression and prognosis of colorectal cancer.
