为了确认Rabbi等2012年发表的木薯遗传图谱9号连锁群与染色体核型的对应关系,本研究利用荧光原位PCR技术将9号连锁群两端的NS701、NS272标记定位到9号染色体上。结果表明,这两个标记均能在华南6号木薯不同分裂期的细胞上检测到1个信号,对其中期细胞进行核型分析,依次将NS701、NS272两个标记分别定位到9号染色体短臂与长臂上,两个标记到着丝粒的百分距离分别为70.66与30.11。NS701、NS272标记均位于木薯9号染色体两端,进一步证实9号连锁群对应9号染色体,本研究为木薯其他连锁群与染色体核型的整合研究奠定了一定的分子细胞遗传学基础。
In order to confirm the correspondence between No 9 linkage group and chromosome karyotype in cassava genetic map published by Rabbi et al. In 2012, we used fluorescence in situ PCR to map the NS701 and NS272 markers located on the 9th linkage group on chromosome 9. The results showed that both of these two markers could detect a signal on the cells of different stage of division in South China cassava No. 6 and analyze the metaphase cells for karyotype analysis. The two markers of NS701 and NS272 were mapped to chromosome 9 short On the arm and on the long arm, the percentages of the two markers to centromeres are 70.66 and 30.11, respectively. The NS701 and NS272 markers were located at both ends of chromosome 9 in Cassava, further confirming that the 9 linkage group corresponds to chromosome 9. This study laid a foundation of molecular cytogenetics for the integrated study of other linkage groups and chromosome karyotype in cassava.
