目的观察早期自然流产患者蜕膜组织中Granulysin、Perforin的表达及其在介导细胞凋亡中的相关性。方法采用免疫组织化学SP法检测30例早期自然流产(流产组)和30例人工流产(对照组)蜕膜组织中Granulysin、Perforin的表达情况,应用TUNEL法检测两组细胞凋亡指数。结果①流产组中Granulysin及Perforin的阳性表达均显著高于对照组,差异有统计学意义(P<0.05)。②Granulysin与Perforin在流产组蜕膜组织中的阳性表达呈正相关(r=0.592,P<0.05)。③流产组的平均细胞凋亡指数[(7.39±3.13)%]高于对照组[(3.45±1.35)%],差异有统计学意义(P<0.05)。④流产组中,Granulysin、Perforin阳性表达组的平均细胞凋亡指数[(8.48±3.08)%,(8.15±3.02)%]均高于阴性表达组[(4.83±1.17)%,(5.00±1.93)%],差异有统计学意义(P<0.05)。结论 Granulysin、Perforin与早期自然流产有关,且两者在早期自然流产的发病过程中有协同作用。
Objective To observe the expression of Granulysin and Perforin in decidual tissue of early spontaneous abortion and its correlation with apoptosis. Methods The expression of Granulysin and Perforin in 30 cases of spontaneous abortion (abortion group) and 30 cases of abortion (control group) deciduas were detected by immunohistochemical SP method. The apoptosis index of the two groups were detected by TUNEL method. Results ① The positive expression of Granulysin and Perforin in abortion group were significantly higher than those in control group (P <0.05). ② The positive expression of Granulysin and Perforin in the decidua of abortion group was positively correlated (r = 0.592, P <0.05). ③ The average apoptosis index in abortion group [(7.39 ± 3.13)%] was higher than that in control group [(3.45 ± 1.35)%], the difference was statistically significant (P <0.05). (4) In the abortion group, the average apoptosis index of Granulysin and Perforin positive groups were significantly higher than that of the negative expression group [(8.48 ± 3.08)% vs (8.15 ± 3.02)% vs (4.83 ± 1.17)% vs (5.00 ± 1.93 )%], The difference was statistically significant (P <0.05). Conclusions Granulysin and Perforin are associated with early spontaneous abortion, and both have synergistic effects in the pathogenesis of early spontaneous abortion.
