目的 :探讨不同放疗剂量对人唾液腺腺样囊性癌(SACC-83、SACC-LM)细胞生物学行为的影响。方法 :采用不同放疗剂量(0、2、4、6、8、10 Gy)作用SACC-83、SACC-LM细胞并继续培养48 h,CCK-8检测细胞存活,流式细胞仪检测细胞凋亡和细胞周期,细胞划痕实验观察细胞迁移能力变化。采用SPSS19.0软件包对数据进行统计学分析。结果:放疗对SACC-LM细胞的细胞存活率、细胞凋亡率、异倍体、细胞迁移力的影响较SACC-83显著(P<0.05);与其他放疗剂量相比,SACC-83细胞在6 Gy剂量照射下细胞存活率最低,细胞凋亡比例及异倍体比例最高,细胞迁移能力最弱。结论:SACC-LM细胞放疗敏感性较SACC-83更高,SACC-83细胞对6 Gy剂量放疗最为敏感。
Objective: To investigate the effects of different doses of radiotherapy on the biological behavior of human salivary adenoid cystic carcinoma (SACC-83, SACC-LM). Methods: SACC-83 and SACC-LM cells were treated with different doses of radiotherapy (0, 2, 4, 6, 8 and 10 Gy) for 48 h. Cell viability was detected by CCK-8 and apoptosis was detected by flow cytometry Cell cycle and cell scratch assay were used to observe the changes of cell migration ability. SPSS19.0 software package for statistical analysis of the data. Results: Compared with SACC-83, SACC-83 cells showed significantly higher cell viability, apoptosis rate, aneuploidy and cell migration than those of SACC-83 Under 6 Gy irradiation, the cell survival rate was the lowest, the proportion of apoptosis and aneuploidy were the highest, and the cell migration was the weakest. CONCLUSIONS: SACC-LM cells are more sensitive to radiotherapy than SACC-83 and SACC-83 cells are most sensitive to 6-Gy radiation.
