目的观察葡萄糖对人脐静脉内皮细胞(HUVECs)活性功能的影响及机制研究。方法内皮细胞培养液体外培养HUVEC,用不同浓度(10、20、30、40、50mmol/L)的葡萄糖作用于HUVEC,用NO含量、eNOS活性检测、细胞凋亡测定和细胞内ROS检测方法,观察不同浓度葡萄糖作用后的HUVEC功能活性的影响。结果高糖对HUVEC的增殖活性具有剂量依赖性抑制作用。高糖剂量依赖性抑制HUVEC细胞eNOs活性、NO的产生,增强细胞凋亡和细胞内ROS的生成。结论葡萄糖对人脐静脉内皮细胞(HUVECs)活性功能具有抑制作用,其机制与增强细胞凋亡和增加细胞内ROS有关。
Objective To observe the effect of glucose on the activity of human umbilical vein endothelial cells (HUVECs) and its mechanism. Methods HUVECs were cultured in vitro with different concentrations of glucose (10, 20, 30, 40 and 50 mmol / L) on HUVECs. HUVECs were treated with NO, eNOS activity, apoptosis and ROS detection. The effects of different concentrations of glucose on the functional activity of HUVECs were observed. Results High glucose had a dose-dependent inhibitory effect on the proliferation of HUVECs. High glucose dose-dependently inhibited the eNOS activity of HUVEC cells, NO production, enhanced apoptosis and intracellular ROS production. Conclusion Glucose has an inhibitory effect on the activity of human umbilical vein endothelial cells (HUVECs), and its mechanism is related to the enhancement of apoptosis and the increase of intracellular ROS.
