目的建立电感耦合等离子体发射光谱测定母乳中钙磷含量的方法,并对深圳市当地母乳钙、磷两种元素的含量进行测定。方法于2011-2013年收集深圳市产后1~2个月产妇的母乳样品共221例,样品冻干保存,经硝酸高压消解后采用电感耦合等离子体发射光谱法(ICP-OES)测定母乳中钙和磷的含量。结果钙检出限0.09 mg/kg,磷检出限0.03 mg/kg;各元素线性良好,相关系数R>0.9999;钙测定精密度1.3%~2.5%,磷测定精密度1.6%~2.1%;国家标准参考物质的分析结果在证书值范围内;深圳市母乳中钙含量187.6~449.6 mg/kg,平均值为294.5 mg/kg,磷含量84.9~230.7 mg/kg,平均值为157.7 mg/kg,钙磷比1.1~3.7,平均值为1.9。结论电感耦合等离子体发射光谱法灵敏、准确可靠,可用于母乳中钙磷元素的同时检测。深圳市母乳中钙元素的平均含量在WHO提供的参考值范围内,磷元素的平均含量略高于WHO提供的参考值范围。
Objective To establish a method for the determination of calcium and phosphorus in breast milk by inductively coupled plasma-atomic emission spectrometry and to determine the contents of calcium and phosphorus in breast milk from Shenzhen. Methods A total of 221 breast-milk samples from 1 to 2 months after delivery in Shenzhen were collected from 2011 to 2013. The samples were freeze-dried and stored. The content of calcium in breast milk was determined by inductively coupled plasma-atomic emission spectrometry (ICP-OES) And phosphorus content. Results The detection limit of calcium was 0.09 mg / kg and the limit of detection of phosphorus was 0.03 mg / kg. The linearity of each element was good with the correlation coefficient R> 0.9999. The precision of determination of calcium was 1.3% ~ 2.5% and the precision of phosphorus determination was 1.6% ~ 2.1%. The results of the national standard reference substance were within the certificate value range. The calcium content of breast milk in Shenzhen was 187.6- 449.6 mg / kg with an average of 294.5 mg / kg and phosphorus content of 84.9-230.7 mg / kg with an average of 157.7 mg / kg , Calcium phosphate ratio of 1.1 to 3.7, an average of 1.9. Conclusion Inductively coupled plasma-atomic emission spectrometry is sensitive, accurate and reliable and can be used for the simultaneous detection of calcium and phosphorus in breast milk. The average content of calcium in breast milk in Shenzhen is within the reference range provided by WHO, and the average content of phosphorus is slightly higher than the reference value provided by WHO.
