目的探讨炎症细胞因子、肝细胞间黏附分子(ICAM-1)在内毒素急性肝损伤发病中的作用机制。方法将 Wistar大鼠随机分成2组:①假手术组(S);②内毒素组(LPS),测定各组大鼠血浆 TNF-α、IL-6水平,肝 ICAM-1免疫组织化学染色及图像分析,肝组织病理观察。结果 LPS 组血浆 TNF-α,IL-6水平和肝血管内皮细胞 ICAM-1表达明显高于 S 组。病理损害较 S 组严重。结论 TNF-α、IL-6及肝 ICAM-1在内毒素急性肝损伤发病机制中起重要作用。
Objective To investigate the mechanism of inflammatory cytokines and intercellular adhesion molecule-1 (ICAM-1) in the pathogenesis of acute liver injury induced by endotoxin. Methods Wistar rats were randomly divided into 2 groups: ① sham operation group (S); ② endotoxin group (LPS), the levels of TNF-α and IL-6 in plasma and the expression of ICAM-1 in liver tissue were detected by immunohistochemistry Image analysis, liver histopathology. Results The levels of TNF-α, IL-6 in plasma and the expression of ICAM-1 in hepatic vascular endothelial cells in LPS group were significantly higher than those in S group. Pathological damage than the S group serious. Conclusion TNF-α, IL-6 and ICAM-1 play an important role in the pathogenesis of acute liver injury induced by endotoxin.
