目的:探讨糖尿病心肌梗死大鼠心肌TGF-β1和CTGF表达的变化以及肝细胞生长因子(HGF)对其影响。方法:6周龄Wistar大鼠90只随机分为5组:对照纽、糖尿病假手术纽、心肌梗死组、糖尿病心梗组和HGF治疗组,每组各18只。以高糖高脂饲料+腹腔注射链尿佐菌素(STZ)诱导建立2型糖尿病大鼠模型后再建立急性心肌梗死(AMI)模型。糖尿病模型大鼠成模后次日,HGF治疗组每天皮下注射HGF,分别观察4周及8周,检测各组心肌组织TGF-β1、CTGF的mRNA及蛋白表达情况。结果:对照组大鼠的TGF-β1、CTGF mRNA及蛋白水平表达微弱;糖尿病心梗组4周后TGF-β1、CTGF mRNA及蛋白水平表达明显升高(P<0.05),8周后表达进一步升高(P<0.01);给予HGF治疗4周后,可以部分降低TGF-β1、CTGFmRNA及蛋白水平的表达(P<0.05);给予HGF治疗8周后,可以明显降低TGF-β1、CTGF mRNA及蛋白水平的表达(P<0.01)。结论:糖尿病心肌梗死大鼠心肌TGF-β1、CTGF的表达明显增高,HGF治疗后能够降低TGF-β1、CTGF的表达,对糖尿病心肌梗死后心肌纤维化的发生有抑制作用。
Objective: To investigate the changes of the expression of TGF-β1 and CTGF and the effect of hepatocyte growth factor (HGF) on myocardial infarction in diabetic rats. Methods: Ninety Wistar rats of 6 weeks old were randomly divided into 5 groups: control group, diabetic neoadjuvant group, myocardial infarction group, diabetes mellitus group and HGF treatment group, 18 rats in each group. Acute myocardial infarction (AMI) model was induced by high glucose and high fat diet + intraperitoneal injection of streptozotocin (STZ) in rats with type 2 diabetes mellitus. The rats in HGF group were injected subcutaneously with HGF daily for 4 and 8 weeks respectively, and the mRNA and protein expression of TGF-β1 and CTGF in each group were detected. Results: The mRNA and protein expression of TGF-β1 and CTGF in the control group were weakly expressed. The expression of TGF-β1, CTGF mRNA and protein in diabetic MI group was significantly increased after 4 weeks (P <0.05) (P <0.01). After treated with HGF for 4 weeks, the expression of TGF-β1 and CTGF mRNA and protein could be partially decreased (P <0.05). After treated with HGF for 8 weeks, the expression of TGF-β1 and CTGF mRNA And protein expression (P <0.01). CONCLUSION: The expression of TGF-β1 and CTGF in myocardium of diabetic rats with myocardial infarction is obviously increased. After HGF treatment, the expression of TGF-β1 and CTGF can be decreased and the myocardial fibrosis can be inhibited after diabetic myocardial infarction.
