我们从黑胸大蠊的若虫中,首次分离发现黑胸大蠊的浓核病毒(PfDNV)。根据Kock’s 定理,将它进行了回感试验,结果表明,对该虫可引起同样的病症,并从中重新分离到完全一样的 DNV。该病毒的回感力也很强,虽然 LT_(50)为39天,但最终死亡率高达99%以上。说明该病毒确系黑胸大蠊若虫高毒力的致死病原体。同时,对该病毒进行了分类鉴定,结果表明,其病毒粒子的形态结构,呈球状二十面体,大小一致,直径为20nm;沉降系数为102s,浮密度为1.4g/cm~3;病毒核酸呈线状,长度为1.7μ,分子量为1.7×10~6d(ssDNV),基因组为单链 DNA;结构蛋白为4条多肽,分子量分别为84K、64K、55K、29K。根据病毒分类命名原则,证明从黑胸大蠊中分离出的病毒属昆虫细小病毒科(Parvoviridae)中的浓核病毒属(Densovirus,DNV),我们将它命名为黑胸大蠊浓核病毒(Periplaneta fuliginosa Densovirus,简称 PfDNV)。PfDNV 的这一初步研究结果,不仅国内外首次发现这一新病毒,且将为开发利用这一资源提供了理论依据和技术途径。
For the first time, we isolated the nodular nucleopolyhedrovirus (PfDNV) from the black-billed Blattella. The genome is single-stranded DNA. The structural protein is 4 peptides with molecular weights of 84K, 64K, 55K and 29K, respectively. According to the principle of virus classification and nomenclature, it was proved that the virus isolated from the black-bai cockatoo belongs to the genus Densovirus (DNV) in the family Parvoviridae and we named it black-spotted cockroach densovirus Periplaneta fuliginosa Densovirus, referred to as PfDNV). The result of this preliminary study of PfDNV not only found this new virus for the first time at home and abroad, but also provided theoretical basis and technical approach for the development and utilization of this resource.
