目的观察益肾平肝方对SHR大鼠的血压及血清神经肽Y(NPY)、降钙素基因相关肽(CGRP)、内皮素(ET)的影响,探讨其在治疗高血压中的降血压机制。方法将21只12周龄雄性自发性高血压大鼠(SHR)随机均分为3组,即益肾平肝方组(YPR组)、贝那普利组(Bena组)、SHR组,另以同周龄7只WKY大鼠作为正常对照组(WKY组)。各组给予相应干预措施12周后,采用尾套法测量大鼠血压,用酶联免疫吸附剂测定法(ELISA)检测大鼠血清NPY、CGRP、ET的含量。结果 SHR组大鼠较WKY组收缩压明显升高(P<0.01)。给予治疗4周、8周、12周后,益肾平肝方组大鼠血压较SHR组明显下降(P<0.01);与SHR组比较,益肾平肝方组大鼠血清NPY、ET的表达量降低,CGRP表达量增加(P<0.01),且与贝那普利组比较差异无统计学意义(P>0.05)。结论益肾平肝方可明显降低SHR大鼠收缩压,其机制可能与抑制NPY、ET的表达及增强CGRP的表达相关。
Objective To observe the effect of Yishen Pinggan decoction on blood pressure, serum neuropeptide Y (NPY), calcitonin gene related peptide (CGRP) and endothelin (ET) in SHR rats and to investigate its antihypertensive effect mechanism. Methods Twenty-one male SHRs of 12 weeks old were randomly divided into three groups: YR group, Bena group, SHR group, Seven WKY rats of the same age were used as normal control group (WKY group). Rats in each group were given corresponding interventions for 12 weeks. The blood pressure of rats was measured by tail cuff method. The contents of NPY, CGRP and ET in serum were detected by enzyme - linked immunosorbent assay (ELISA). Results Compared with WKY group, systolic pressure in SHR group was significantly increased (P <0.01). After 4 weeks, 8 weeks and 12 weeks of treatment, the blood pressure of Yishenpinggan decoction group was significantly lower than that of SHR group (P <0.01). Compared with SHR group, the levels of NPY and ET The expression of CGRP was decreased (P <0.01), and there was no significant difference between the two groups (P> 0.05). Conclusion Yishen Pinggan Decoction can significantly reduce systolic blood pressure in SHR rats, and its mechanism may be related to the inhibition of the expression of NPY and ET and the enhancement of CGRP expression.
