目的:研究淋巴细胞中端粒保护蛋白(POT1)的表达、端粒酶活性与2型糖尿病发生发展的关系。方法:以2型糖尿病患者162例为实验组,以健康体检者162例为对照组,RT-PCR检测POT1mRNA的表达;TRAP-ELISA分析端粒酶活性。结果:实验组POT1 mRNA的表达和端粒酶活性均明显高于对照组。其中血糖控制尚可组和差组分别与对照组比较,POT1 mRNA表达和端粒酶活性均明显增高;按糖尿病性视网膜病变分期中各组POT1mRNA表达和端粒酶活性与对照组比较存在显著性差异;且病程5~10年组、10年以上组与5年以下组相比,POT1mRNA表达和端粒酶活性也存在显著性差异。结论:2型糖尿病淋巴细胞中POT1mRNA的表达和端粒酶活性均明显增高,且与病情发展呈正相关。POT1和端粒酶的表达变化作为预后的评价依据有重要的临床意义。
Objective: To study the relationship between the expression of Telomere protective protein (POT1), telomerase activity and the development of type 2 diabetes mellitus in lymphocytes. Methods: Totally 162 patients with type 2 diabetes mellitus (T2DM) were selected as experimental group and 162 healthy controls as control group. The expression of POT1 mRNA was detected by RT-PCR and telomerase activity was analyzed by TRAP-ELISA. Results: The expression of POT1 mRNA and telomerase activity in experimental group were significantly higher than those in control group. POT1 mRNA expression and telomerase activity were significantly higher in the control group and the control group than in the control group and the control group (P <0.01). The expression of POT1 mRNA and telomerase activity in each group were significantly higher than those in the control group according to the stage of diabetic retinopathy There was also a significant difference in POT1mRNA expression and telomerase activity between the 5 ~ 10 year group and the 10 - year group and the 5 - year group. Conclusion: The expression of POT1 mRNA and telomerase activity in type 2 diabetic lymphocytes were significantly increased, and positively correlated with the progression of the disease. POT1 and telomerase expression changes as a prognostic evaluation based on important clinical significance.
