目的观察参附注射液对脓毒症大鼠心血管功能和心肌氧化应激的改善作用。方法采用结扎穿孔盲肠法制备腹腔感染脓毒症动物模型。将180只SD大鼠按随机数字表法分为假手术(Sham)组、脓毒症(CLP)组、参附注射液低剂量(LSF)组、参附注射液高剂量(HSF)组,每组各20只。记录手术后6、18 h各组的平均动脉压(MAP)、心率(HR)、左室收缩压(LVDP)和左室最大收缩速率(+dP/dt)、左室舒张压(LVEDP)和左室最大舒张速率(-dP/dt)、心肌细胞凋亡发生率(AI)、心脏组织丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、呼吸控制比(RCR)。结果与Sham组比较,CLP组术后6h时的HR显著升高(P<0.05),但术后18h时的HR有所降低(P<0.01);术后6、18h的MAP、LVDP、+dP/dt、LVEDP、dP/dt、SOD、RCR均显著下降(均P<0.01),AI、MDA均显著升高(P<0.05或0.01)。而与CLP组比较,LSF组术后6h的MAP和HR、HSF组术后6h的HR差异均无统计学意义(均P>0.05),但HSF组术后6h的MAP及LSF、HSF组术后18h的HR、MAP均明显升高(P<0.05或0.01);LSF组和HSF组术后6、18h的LVDP、+dP/dt、LVEDP、-dP/dt、SOD、RCR均显著升高(P<0.05或0.01);LSF组和HSF组术后6、18h的AI、MDA均显著降低(P<0.05或0.01)。结论参附注射液可改善脓毒症大鼠的左室收缩和舒张功能,减少心肌细胞凋亡和心肌组织MDA含量,增加SOD活性,保护线粒体功能,且这些作用的强度与参附注射液药物剂量成正比。
Objective To observe the effect of Shenfu injection on cardiovascular function and myocardial oxidative stress in septic rats. Methods The peritoneal sepsis model was established by ligation and puncture of the cecum. 180 SD rats were randomly divided into Sham group, CLP group, LSF group, HSF group, Each group of 20. Conclusion Shenfu injection can improve left ventricular systolic and diastolic function, reduce myocardial cell apoptosis and myocardial MDA content, increase SOD activity and protect mitochondrial function in septic rats, and the intensity of these effects is similar to that of Shenfu Injection The dose is proportional.
