目的:建立大鼠PDLLA/FK506(外消旋聚乳酸/FK506)复合套管坐骨神经再生模型,观察大鼠坐骨神经再生过程中行为学、痛阈以及脊髓背角c-fos表达的改变,研究大鼠PDLLA/FK506复合套管外周神经再生模型中神经病理痛的变化.方法:雄性Wistar大鼠40只,体质量180~200g,随机分成2组,每组20只,A组为坐骨神经损伤PDLLA/FK506套管修复组;B组为假手术组.术后27d起检测行为学、机械刺激阈值、热缩足反射时间及c-fos表达计数.结果:A组术后27d后机械痛阈和热缩足反射潜伏期与B组相差明显,有统计学意义(P<0.05).A组术侧L4~5脊髓背角与B组相比较,背角浅层c-fos计数增多,有统计学意义(P<0.05).A组组内c-fos、机械痛阈和热缩足反射潜伏期45d与27d比较均有统计学意义(P<0.05).结论:大鼠PDLLA/FK506复合套管外周神经再生模型中神经再生过程中诱发脊髓背角浅层c-fos表达,同时出现再生神经支配区域的神经病理痛和疼痛相关行为.
OBJECTIVE: To establish a sciatic nerve regeneration model of PDLLA / FK506 composite cannula in rats and observe the changes of behavior, pain threshold and c-fos expression in spinal dorsal horn during rat sciatic nerve regeneration. PDLLA / FK506 composite cannula.Methods: Forty male Wistar rats weighing 180 ~ 200g were randomly divided into two groups (20 rats in each group), group A was sciatic nerve injury PDLLA / FK506 Group B was sham-operation group.The detection behavior, mechanical stimulation threshold, refraction time of reflex foot and c-fos expression count were measured at 27 days after operation.Results: The mechanical pain threshold and thermal contraction There was a significant difference between group B and group B (P <0.05) .Compared with group B, the number of c-fos in superficial dorsal horn of L4-5 spinal cord in group A increased significantly (P <0.05) P <0.05) .A group of c-fos, mechanical pain threshold and thermal reflex foot reflex latency 45d and 27d compared with statistical significance (P <0.05) .Conclusion: PDLLA / FK506 composite sheath of peripheral nerve regeneration In the model, the c-fos expression in the dorsal horn of the spinal cord is induced in the process of nerve regeneration, and the nerves in the regenerating innervation area Physical pain and pain-related behavior.
