目的检测悬浮红细胞保存期末的质量情况。方法无菌接驳取悬浮红细胞标本,分别在保存前(0 d)和保存期末(35 d)进行红细胞压积(HCT)、血红蛋白(HGB)、红细胞计数(RBC)和游离血红蛋白检测,并使用透射电镜观察保存前和保存期末细胞形态结构的变化。结果在保存前(0 d)和保存期末(35 d)悬浮红细胞HCT、HGB、RBC等指标均符合质量要求,悬浮红细胞在保存前和保存期末HCT、HGB、RBC等指标变化不大,差异均无统计学意义(P>0.05);保存35 d后,红细胞游离血红蛋白浓度增高为(149.5±9.6)mg/L,与保存前的(26.5±1.9)mg/L比较,差异有统计学意义(P<0.05);透射电镜下观察其形态无明显变化,红细胞为双凹圆盘状,细胞膜结构清楚,膜表面光滑完整,未见突起、凹陷、空泡、缺失等膜损伤。结论悬浮红细胞保存前和保存期末各项检测指标均符合质量要求;控制悬浮红细胞制备和保存中各个环节,对于保护红细胞的形态和功能,保证血液质量,保证血液输注效果,均具有重要意义。
Objective To test the quality of suspended erythrocytes at the end of storage. Methods Aseptic connections of suspended erythrocyte samples were performed. HCT, HGB, RBC and free hemoglobin were detected before and at the end of storage (0 d) and The changes of cell morphology before and at the end of storage were observed by transmission electron microscope. Results The indexes of HCT, HGB, RBC and so on were all in line with the quality requirements before and at the end of the storage (0 d) and 35 d. The indexes of HCT, HGB and RBC before and after the storage (P> 0.05). After 35 days of storage, the free hemoglobin concentration in erythrocytes increased to (149.5 ± 9.6) mg / L, which was significantly lower than that before storage (26.5 ± 1.9) mg / L P <0.05). The morphology of the erythrocytes did not change obviously under the transmission electron microscope. The erythrocytes were biconcave, the membrane structure of the erythrocytes was clear, the membrane surface was smooth and intact, and there was no membrane damage such as protrusions, depressions, vacuoles and missing. Conclusion The detection indexes before and during the storage period of the suspension red blood cells meet the quality requirements. It is of great significance to control the morphology and function of the erythrocytes, to ensure the blood quality and to ensure the blood transfusion effect.
