目的:利用活体成像技术观察黄芪多糖对裸鼠卵巢癌细胞休眠的维持作用。方法:选取建立成功的卵巢癌细胞休眠裸鼠模型30只随机分成3组:即对照组、黄芪多糖组、顺铂组,每组10只,连续给药14天;利用活体成像系统分别在给药前1天、给药后第7天、第14天监测各组裸鼠卵巢癌细胞休眠情况。结果:与对照组比较,黄芪多糖组给药后第7天、第14天,裸鼠体质量无明显变化;而顺铂组在给药后第7天、第14天,体质量明显降低,差异均有统计学意义(P<0.01)。对照组裸鼠卵巢癌休眠细胞光子量随着时间而升高。与对照组比较,黄芪多糖组和顺铂组在给药第7天、第14天时,裸鼠肿瘤休眠细胞光子量明显降低,差异均有统计学意义(P<0.05,P<0.01)。结论:黄芪多糖具有较好的维持裸鼠卵巢癌细胞休眠的作用,活体成像技术可动态地观察肿瘤细胞休眠的情况,为黄芪多糖应用于临床提供理论依据。
Objective: To observe the effect of Astragalus Polysaccharide on the maintenance of ovarian cancer cell in nude mice by using live imaging technique. Methods: Totally 30 ovarian cancer-bearing nude mice models were randomly divided into three groups: control group, Astragalus polysaccharide group and cisplatin group, with 10 mice in each group. The mice were continuously administered for 14 days. Using live imaging system, One day before the drug administration, on the 7th day and the 14th day after administration, the ovarian cancer cells in each group were monitored for dormancy. Results: Compared with the control group, there was no significant change in the body weight of the nude mice on the 7th day and the 14th day after administration of the APS group. On the 7th and 14th day after administration, the body weight of the APS group was significantly decreased, The differences were statistically significant (P <0.01). The photon quantity of dormant cells in control group nude mice with ovarian cancer increased with time. Compared with the control group, phototherapy of APS in cisplatin group and APS group on the 7th day and the 14th day were significantly decreased (P <0.05, P <0.01). Conclusion: Astragalus polysaccharide has a good effect on maintaining the ovarian cancer cells in nude mice. Live imaging technique can dynamically observe the dormancy of tumor cells and provide a theoretical basis for the clinical application of astragalus polysaccharide.
