目的:研究健骨固本胶囊对维甲酸所致大鼠实验性骨质疏松症的影响及可能作用机制。方法:通过灌胃维甲酸75mg·kg-1·d-1,连续2周复制大鼠骨质疏松模型,同时给予药物干预4周后,使用双能X线骨密度测定仪测定全身骨矿量(BMC)和全身骨密度(BMD)及股骨BMD;采用市售试剂盒分析血清抗酒石酸酸性磷酸酶(StrACP)活力和骨组织羟脯氨酸(HOP)含量;股骨三点弯曲实验分析最大应力、弹性模量和弹性能量吸收。结果:健骨固本胶囊可显著增加全身BMC,全身及股骨BMD;增加股骨干重指数;显著降低血清StrACP水平,提高骨HOP含量;提高骨最大应力、弹性模量和弹性能量吸收。结论:健骨固本胶囊对维甲酸所致大鼠实验性骨质疏松症模型具有显著的改善作用。
Objective: To study the effect of Jiangu Guben Capsule on experimental osteoporosis induced by retinoic acid and its possible mechanism. METHODS: Rat model of osteoporosis was duplicated by intragastric administration of retinoic acid 75 mg·kg-1·d-1 for 2 weeks. After 4 weeks of drug intervention, the amount of whole body bone mineral was measured by dual energy X-ray absorptiometry. (BMC) and whole body bone density (BMD) and femur BMD; serum anti tartaric acid phosphatase (StrACP) activity and bone tissue hydroxyproline (HOP) content were analyzed using commercially available kits; femoral three-point bending test was used to analyze the maximum stress , elastic modulus and elastic energy absorption. RESULTS: Jiangu Guben Capsules can significantly increase BMD of whole body and femur, increase femoral dry weight index, significantly decrease serum StrACP level, increase bone HOP content, and increase bone maximum stress, elastic modulus, and elastic energy absorption. Conclusion: Jiangu Guben Capsules can significantly improve the experimental osteoporosis model induced by retinoic acid in rats.
