目的:探讨屋尘螨过敏症母亲脐血中CD4+CD25+Foxp3+调节性T(Treg)细胞和CD4+IL-10+调节性T(Treg)细胞是否存在数量上的缺陷。方法:收集7例患屋尘螨过敏症及14例无过敏症母亲的脐血,分离脐血单个核细胞(CBMC)后体外培养,分别用及不用天然屋尘螨抗原1(nDer p1)刺激,用流式细胞术检测刺激及不刺激时CD4+CD25+Foxp3+Treg细胞及CD4+IL-10+Treg细胞占CD4+T细胞的比率。结果:不刺激时,过敏症母亲组CBMC中的CD4+CD25+Foxp3+Treg细胞占CD4+T细胞的比率为(1.13±0.36)%,显著低于无过敏症母亲组的比率(3.11±1.13)%(P<0.01)。尘螨抗原刺激后,两组CD4+CD25+Foxp3+Treg细胞比率均较不刺激时升高(P<0.01),过敏症母亲组增加的比率为(0.67±0.25)%,低于无过敏症母亲组增加的比率(1.50±0.40)%(P<0.01);不刺激时,过敏症母亲组CD4+IL-10+Treg细胞比率的中位数为0.41%,显著低于无过敏症母亲组的1.04%(P<0.01);尘螨刺激后,无过敏症组CD4+IL-10+Treg细胞比率2.26%较该组不刺激时比率1.04%升高(P<0.05),显著高于过敏症母亲组的比率0.61%(P<0.01),但两组提高的幅度未见统计学差异(P>0.05)。结论:过敏症母亲脐血中CD4+CD25+Foxp3+Treg细胞及CD4+IL-10+Treg细胞在数量上存在缺陷。
Objective: To investigate whether there is a quantitative defect in CD4 + CD25 + Foxp3 + Treg cells and CD4 + IL-10 + Treg cells in umbilical cord blood of house-dust mite allergic mothers. Methods: Cord blood samples from 7 house dust mite allergy patients and 14 allergic mothers were collected and cultured in vitro. Cord blood mononuclear cells (CBMC) were isolated and cultured with and without nDer p1 The percentage of CD4 + CD25 + Foxp3 + Treg cells and CD4 + IL-10 + Treg cells in CD4 + T cells were measured by flow cytometry. Results: The ratio of CD4 + CD25 + Foxp3 + Tregs to CD4 + T cells in CBMC of allergic mothers was (1.13 ± 0.36)%, significantly lower than that in mothers without allergy (3.11 ± 1.13 )% (P <0.01). The ratio of CD4 + CD25 + Foxp3 + Treg cells was increased in both groups after stimulation with dust mite antigen (P <0.01), and was significantly higher in allergic mothers (0.67 ± 0.25)% than that without allergy (1.50 ± 0.40)% (P <0.01). The median of CD4 + IL-10 + Treg cells in allergic mothers was 0.41%, which was significantly lower than that in non-allergic mothers (P <0.01). The percentage of CD4 + IL-10 + Treg cells in allergy-free group was 2.26% higher than that in non-allergic group The rate of symptomatic mother was 0.61% (P <0.01), but there was no significant difference between the two groups (P> 0.05). Conclusion: CD4 + CD25 + Foxp3 + Treg cells and CD4 + IL-10 + Treg cells in cord blood of allergic mothers are deficient in quantity.
