目的通过荧光活性染料DiI标记观察骨髓基质干细胞在部分脱钙骨上的粘附。方法用DiI标记人骨髓基质干细胞,接种于部分脱钙骨上,测定细胞的粘附率。于接种后第2、4、7天激光共聚焦显微镜观察细胞在支架材料上的生长状况。结果DiI标记的骨髓基质干细胞的粘附率为(99.9±0.2)%,未标记的骨髓基质干细胞粘附率为(99.1±1)%,两者无统计学差异(P>0.05);第7天时细胞脱钙骨内孔覆盖。结论荧光活性染料DiI标记不影响骨髓基质干细胞在部分脱钙骨支架上的粘附,是观察骨髓基质干细胞在脱钙骨支架上粘附状况的较好方法。
Objective To observe the adhesion of bone marrow stromal stem cells on partially decalcified bone by fluorescently labeled DiI. Methods Human bone marrow stromal cells were labeled with DiI and seeded on some decalcified bone to determine the cell adhesion rate. On the 2nd, 4th and 7th days after inoculation, the confocal microscope was used to observe the growth of the cells on the scaffolds. Results The adhesion rate of DiI-labeled bone marrow stromal stem cells was (99.9 ± 0.2)% and that of unlabeled bone marrow stromal stem cells was (99.1 ± 1)%, with no significant difference (P> 0.05) Day when the cells decalcified bone inner covering. Conclusion DiI-labeled fluorescent dye does not affect the adherence of bone marrow stromal cells to partially decalcified bone scaffolds. It is a good way to observe the adhesion of bone marrow stromal cells to decalcified bone scaffolds.
