目的:探讨阿托伐他汀和CoQ10对高胆固醇兔心肌线粒体氧自由基损伤的保护作用。方法:40只健康2月龄雄性新西兰白兔,随机均分为5组:正常对照组(A组)、高脂饮食组(B组)、高脂饮食+阿托伐他汀组(C组)、高脂饮食+阿托伐他汀+低剂量CoQ10组(D组)及高脂饮食+阿托伐他汀+高剂量CoQ 10组(E组)。6周末制备心肌线粒体和电镜标本。探讨各组心肌线粒体中丙二醛和谷胱甘肽的含量及电镜下超微结构的变化。结果:A组与E组心肌线粒体丙二醛含量低于B组(P<0.05),B组谷胱甘肽含量低于A组(P<0.05),E组心肌线粒体谷胱甘肽含量明显高于B组(P<0.01)。电镜下观察:与A组比较,B组心肌细胞超微结构损伤严重;E组心肌细胞超微结构保持良好,接近A组;C组与D组心肌细胞超微结构有不同程度的损伤,介于上述两组之间。结论:阿托伐他汀与高剂量CoQ10合用可降低心肌线粒体中的丙二醛,增加谷胱甘肽含量,保护心肌线粒体免受脂质过氧化损伤,达到保护心肌细胞的目的。
AIM: To investigate the protective effects of atorvastatin and CoQ10 on myocardial oxygen free radical damage in hypercholesterolemic rabbit hearts. Methods: Forty healthy male New Zealand white rabbits aged 2 months were randomly divided into 5 groups: normal control group (group A), high fat diet group (group B), high fat diet group and atorvastatin group (group C) , High fat diet + atorvastatin + low dose CoQ10 group (D group) and high fat diet + atorvastatin + high dose CoQ 10 group (E group). Myocardial mitochondria and electron microscopy specimens were prepared at 6 weeks. To investigate the contents of malondialdehyde and glutathione in myocardial mitochondria and the ultrastructural changes under electron microscope. Results: The contents of MDA in group A and group E were lower than those in group B (P <0.05), while those in group B were lower than those in group A (P <0.05). The content of glutathione in myocardial mitochondria of group E was significantly Higher than B group (P <0.01). Electron microscopic observation: Compared with group A, myocardial ultrastructure of group B was severely damaged. The ultrastructure of myocardial cells in group E remained well, which was close to group A. The ultrastructure of myocardial cells in groups C and D were damaged to some extent, Between the above two groups. Conclusions: Combination of atorvastatin and high dose of CoQ10 can reduce myocardial malondialdehyde, increase glutathione content, protect myocardial mitochondria from lipid peroxidation injury, and achieve the purpose of protecting cardiomyocytes.
