目的观察凉血通瘀方对H2O2诱导损伤的血管内皮细胞增殖、凋亡及SOD表达的影响,探讨凉血通瘀方治疗出血性中风的可能机理。方法 100μmol/LH2O2造成血管内皮细胞氧化应激损伤模型,采用MTT法测定阴性对照组,模型组,凉血通瘀方高(2g/mL)、中(1g/mL)、低(0.5g/mL)剂量组细胞活性和细胞凋亡情况,黄嘌呤氧化酶法检测各组细胞上清液中SOD的活力。结果 100μmol/LH2O2可抑制血管内皮细胞增殖,促进细胞凋亡,降低SOD活力;而凉血通瘀方则能提高H2O2诱导损伤的血管内皮细胞活性,减轻内皮细胞的凋亡,提升SOD活力。这可能是凉血通瘀方治疗出血性中风机理所在。结论凉血通瘀方可能通过抗氧化和减轻内皮细胞的凋亡,发挥其对血管内皮细胞的保护作用。
Objective To observe the effects of Liangxuetongyu Decoction on proliferation, apoptosis and SOD expression of vascular endothelial cells induced by H2O2, and to explore the possible mechanism of Liangxuetongyu Decoction in treating hemorrhagic stroke. Methods 100μmol / LH 2 O 2 induced the oxidative stress injury of vascular endothelial cells. MTT assay was used to detect the oxidative stress in the model group, the model group, Lianggutongyu et al. (2g / mL) ) Dose group of cell activity and apoptosis, xanthine oxidase method was used to detect the superoxide dismutase activity in each group. Results 100μmol / LH 2 O 2 could inhibit the proliferation of vascular endothelial cells, promote cell apoptosis and decrease the activity of SOD. However, Liangxuetongyu Decoction could increase the activity of vascular endothelial cells induced by H2O2, reduce the apoptosis of endothelial cells and increase the activity of SOD. This may be the treatment of hemorrhagic stroke of cooling blood stasis side. Conclusion Liangxuetongyu Decoction may exert its protective effect on vascular endothelial cells through anti-oxidation and reducing apoptosis of endothelial cells.
