目的选取表皮生长因子受体(EGFR)低表达的子宫内膜癌Ishikawa细胞和经转染过表达EGFR的Ishikawa-p LWERNL细胞,研究EGFR对子宫内膜癌细胞迁移能力和侵袭能力的影响。方法体外迁移试验比较转染前后2种子宫内膜癌细胞的迁移能力。体外侵袭试验检测转染前后2种子宫内膜癌细胞的侵袭能力。蛋白印迹法比较转染前后2种子宫内膜癌细胞基质金属蛋白MMP9、MMP2表达水平的差异。结果和转染前的Ishikawa细胞相比较,经转染稳定过表达EGFR的Ishikawa-p LWERNL细胞迁移能力更强,差异有统计学意义(t=6.148,P<0.01)。转染后,稳定过表达EGFR的Ishikawa-p LWERNL细胞侵袭能力增强,差异有统计学意义(t=10.028,P<0.01)。和EGFR低表达的Ishikawa细胞相比较,EGFR高表达的Ishikawa-p LWERNL细胞基质金属蛋白酶MMP9、MMP2的表达水平较高,差异有统计学意义(t值分别为6.934,5.748,P值均<0.05)。结论 EGFR过表达可增强子宫内膜癌细胞的迁移能力和侵袭能力。
Objective To investigate the effect of EGFR on the migration and invasion of endometrial carcinoma cells by selecting Ishikawa cells with low expression of epidermal growth factor receptor (EGFR) and Ishikawa-p LWERNL cells transfected with EGFR. Methods Migration in vitro was compared between two endometrial cancer cells before and after transfection. In Vitro Invasion Assay The invasion ability of 2 endometrial cancer cells before and after transfection was tested. Western blotting was used to compare the expression of matrix metalloproteinases MMP9 and MMP2 in the two kinds of endometrial carcinoma cells before and after transfection. Results Compared with Ishikawa cells before transfection, Ishikawa-p LWERNL cells stably overexpressed EGFR had stronger migration ability (t = 6.148, P <0.01). After transfection, the invasion ability of Ishikawa-p LWERNL cells stably overexpressing EGFR was enhanced, with a significant difference (t = 10.028, P <0.01). Compared with Ishikawa cells with low expression of EGFR, the expression of matrix metalloproteinases MMP9 and MMP2 in Ishikawa-p LWERNL cells with high EGFR expression was significantly higher (t = 6.934,5.748, P <0.05) ). Conclusion Overexpression of EGFR can enhance the migration and invasion of endometrial cancer cells.
