以浙江雁荡山×云南广南种质的杂交组合茎段为外植体,筛选出1/2MS+IBA 1.0 mg.L-1+6-BA 1.0 mg.L-1腋芽诱导培养基与1/2MS+IBA 1.5 mg.L-1+6-BA 0.5 mg.L-1芽增殖培养基,在浙江、云南、广西、湖南等159个铁皮石斛野生与人工种质中应用,腋芽诱导率达94.3%,具有较好的通用性;芽增殖培养时,不同种质分化出单芽、丛芽、原球茎3种增殖途径,增殖效果存在显著差异。研究结果表明,不同种质铁皮石斛的增殖对培养基有专用性需求,开发铁皮石斛无性系繁育专用培养基具有紧迫性和重要的应用价值。
The stems of hybrids of Yandang Mountain × YunnanGuangnan germplasm were used as explants to select 1 / 2MS + IBA 1.0 mg.L-1 + 6-BA 1.0 mg.L-1 axillary bud induction medium and 1 / 2MS + IBA 1.5 mg.L-1 + 6-BA 0.5 mg.L-1 bud proliferation medium was used in 159 wild and artificial germplasms of Dendrobium officinale in Zhejiang, Yunnan, Guangxi and Hunan provinces. The induction rate of axillary bud was 94.3 %, Has good versatility; bud propagation culture, different germplasm differentiation of single buds, buds buds, protocorm three proliferation pathway, the proliferation of significant differences exist. The results showed that the proliferation of different varieties of Dendrobium candidum has a special needs of the medium, the development of Dendrobium officinale breeding special medium has the urgency and important application value.
