目的探讨补肾活血方、补肾方、活血方对血管性痴呆小鼠学习记忆能力及脑组织碱性成纤维细胞生长因子(bFGF)mRNA表达的影响。方法采用脑缺血再灌注法复制血管性痴呆小鼠模型,以尼莫地平为阳性对照组,观察补肾方、活血方、补肾活血方3组小鼠水迷宫法行为学、脑组织bFGF含量及bFGF mRNA表达的变化。结果与假手术组比较,模型组小鼠学习与记忆能力下降,表现为游全程时间延长,错误次数增加(P<0.01),脑组织bFGF含量和bFGF mRNA表达相对含量明显升高(P<0.05或P<0.01);与模型组比较,各治疗组学习成绩和记忆成绩均有明显提高(P<0.05或P<0.01),脑组织bFGF含量和bFGF mRNA表达相对含量明显升高(P<0.01);各治疗组比较,补肾活血组的学习成绩与记忆成绩优于其他治疗组(P<0.05),bFGF含量及脑组织bFGF mRNA表达相对含量均高于活血组(P<0.05),与补肾组比较无统计学差异(P>0.05)。结论补肾方、活血方、补肾活血方均可通过促进脑组织bFGF mRNA表达,提高脑组织bFGF含量,改善血管性痴呆小鼠学习记忆能力,且补肾活血方在促进小鼠学习记忆能力方面显著优于补肾方和活血方,在调节bFGF含量及bFGFmRNA表达方面明显优于活血方。
Objective To investigate the effects of Bushen Huoxue Recipe, Bushen Recipe and Huoxue Recipe on the learning and memory abilities and the expression of basic fibroblast growth factor (bFGF) mRNA in mice with vascular dementia. Methods The mouse model of vascular dementia was established by cerebral ischemia-reperfusion. Nimodipine was used as the positive control group. The water maze behavior, the content of bFGF in brain tissues and the content of bFGF in brain tissues of mice in Bushen Fang, Huoxue Fang and Bushen Huoxue were observed. bFGF mRNA expression changes. Results Compared with the sham-operation group, the learning and memory abilities of the model group decreased significantly (P <0.05), the prolongation of the whole journey and the increase of the number of errors (P <0.01), and the relative content of bFGF and bFGF mRNA (P <0.01 or P <0.01). Compared with the model group, the scores of learning and memory in each treatment group were significantly increased (P <0.05 or P <0.01), the content of bFGF and the relative content of bFGF mRNA in brain tissue were significantly increased (P <0.05). The content of bFGF and the relative content of bFGF mRNA in brain tissue of Bushenhuoxue group were higher than those of Huoxuehuoxue group (P <0.05) There was no significant difference between the two groups (P> 0.05). Conclusion Bushenfanghuoxue recipe and Bushenhuoxue prescription can improve bFGF content in brain tissue, improve learning and memory abilities of mice with vascular dementia by promoting bFGF mRNA expression in brain tissue, and Bushenhuoxue Fang is significantly superior in learning and memory ability of mice It is obviously superior to Huoxue Recipe in regulating the content of bFGF and the expression of bFGF mRNA in Bushen Recipe and Huoxue Recipe.
