目的:探讨乳腺癌组织内血管内皮生长因子-C mRNA(VEGF-C mRNA)的表达及微血管密度(MVD),分析MVD和VEGF-C mRNA表达与乳腺癌患者预后的意义。方法:采用原位杂交技术观察92例乳腺浸润性导管癌(IDC)组织中VEGF-C mRNA的表达;CD105(细胞膜糖蛋白)标记乳腺癌组织中微血管并观察MVD。结果:不同组织学分级乳腺IDC组织中VEGF-C mRNA表达差异有统计学意义,P<0.05;在淋巴结转移组VEGF-C mRNA表达明显高于淋巴结未转移组,P<0.01;VEGF-C mRNA表达阳性组MVD明显高于VEGF-C mRNA表达阴性组,P<0.001;淋巴结转移组乳腺癌组织中MVD高于淋巴结未转移组,差异有统计学意义(P<0.001),VEGF-C mRNA表达阳性组(高MVD组)较VEGF-C mRNA表达阴性组(低MVD组)5年生存率低,差异有统计学意义,P<0.05。结论:VEGF-C mRNA表达与乳腺癌淋巴结转移及MVD密切相关,检测乳腺癌组织中VEGF-C mRNA表达及观察MVD可能预测乳腺癌预后。
Objective: To investigate the expression of vascular endothelial growth factor-C mRNA (VEGF-C mRNA) and microvessel density (MVD) in breast cancer and to analyze the significance of MVD and VEGF-C mRNA expression in prognosis of patients with breast cancer. Methods: The expression of VEGF-C mRNA in 92 cases of invasive ductal carcinoma of the breast (IDC) tissues was observed by in situ hybridization. Microvessels in breast cancer tissues were labeled with CD105 (cell membrane glycoprotein) and MVD was observed. Results: The expression of VEGF-C mRNA in IDC tissues with different histological grade had statistical significance (P <0.05). The expression of VEGF-C mRNA in lymph node metastasis group was significantly higher than that in non-metastasis lymph node group (P <0.01) The MVD in positive group was significantly higher than that in negative group (P <0.001). The MVD in lymph node metastasis group was higher than that in lymph node group (P <0.001), and the expression of VEGF-C mRNA The 5-year survival rate of positive group (high MVD group) was lower than that of VEGF-C negative group (low MVD group), the difference was statistically significant (P <0.05). Conclusion: The expression of VEGF-C mRNA is closely related to lymph node metastasis and MVD in breast cancer. Detecting the expression of VEGF-C mRNA in breast cancer tissue and observing MVD may predict the prognosis of breast cancer.
