目的利用动态增强磁共振成像技术DCE-MRI成像参数评价抗血管药物抑制肿瘤血管生成的疗效。方法 48只结肠癌裸鼠移植瘤随机分两组,对照组:24只、抗肿瘤药物组:24只,对照组注射等量的0.9%氯化钠溶液、腹腔注射5-氟尿嘧啶(5-FU)0.02ml/天,分别在治疗前与治疗后2天、7天及14天进行常规MRI检查和动态增强检查,治疗后每个时间段各处死6只老鼠行免疫组化染色,与动态增强检查各参数进行相关性分析。结果在DCE-MRI各参数,实验组Ktrans值及Kep值与对照组差异有统计学意义(P<0.05),同时间点Ktrans值及同时段的Kep值实验组与对照组比较均有统计学意义(P<0.05),并与免疫组化结果做分析,存在良好的相关性。参数值Ve不存在统计学意义。结论裸鼠移植瘤模型提供了一个理想的动物模型研究肿瘤,DCE-MRI成像半定量和定量参数分析肿瘤新生血管生成或抗肿瘤药物治疗后肿瘤血管生成关闭的应用价值。
Objective To evaluate the therapeutic effect of anti-angiogenic agents on tumor angiogenesis by dynamic contrast-enhanced magnetic resonance imaging (DCE-MRI). Methods Forty-eight colon cancer xenografts in nude mice were randomly divided into two groups: control group (n = 24), antineoplastic group (n = 24), control group with 0.9% sodium chloride solution and intraperitoneal injection of 5-fluorouracil ) 0.02ml / day, conventional MRI examination and dynamic contrast enhancement were performed before treatment and 2 days, 7 days and 14 days after treatment respectively. After the treatment, 6 mice sacrificed at each time point for immunohistochemical staining and dynamic enhancement Check the parameters for correlation analysis. Results There was a significant difference between the Ktrans value and the Kep value in the experimental group and the control group (P <0.05), Ktrans value in the same time point and Kep value in the same period in the experimental group compared with the control group Significance (P <0.05), and immunohistochemical analysis of the results, there is a good correlation. The parameter value Ve does not have statistical significance. Conclusion The model of nude mice transplantation provides an ideal animal model for the study of tumor. The value of semi-quantitative and quantitative analysis of DCE-MRI in the analysis of tumor angiogenesis after tumor angiogenesis or anti-tumor drug treatment.
