目的和方法 :采用无血清成年大鼠肺组织培养 ,用液体闪烁计数器测定3H 胆碱掺入磷脂酰胆碱量 ,消化定磷法测总磷脂 ,薄层层析及薄层扫描测磷脂各组分含量变化 ,观察生理浓度表皮生长因子对成年大鼠肺表面活性物质合成的调控。结果 :① 10 -9mol/LEGF作用 8h后 ,PC合成量显著增加 ,16h达高峰 ;②EGF可显著增加总磷脂、PS特征性成分PC、PG合成 (P <0 .0 1)。而细胞膜特征性组分PE、PSe、SM、PI均无显著影响 (P >0 .0 5 ) ;③PTK抑制剂Tyrphostin 47、PKC抑制剂H7均对EGF促PS合成有阻断效应。④EGF、TP47、H7对肺组织LDH释放量无显著影响。结论 :EGF参与生理状态下成年大鼠PS合成的调控 ,其信号转导与EGF受体PTK和细胞内PKC途径有关
PURPOSE AND METHODS: Lung tissue cultures of serum-free adult rats were performed. The amount of phosphatidyl choline incorporated into 3H-choline was determined by liquid scintillation counter. Total phospholipids were determined by digestion and determination of phosphorus. The change of sub-content, observe the physiological concentration of epidermal growth factor on the synthesis of lung surfactant in adult rats. Results: ① After 8 hours of 10 -9 mol / L EGF treatment, the amount of PC synthesis increased significantly and reached a peak at 16 hours; ②EGF increased the synthesis of total phospholipid and PS, PC and PG (P <0.01). (P> 0.05). ③PTK inhibitors Tyrphostin 47 and PKC inhibitor H7 both had a blocking effect on EGF-induced PS synthesis. ④EGF, TP47, H7 had no significant effect on LDH release from lung tissue. CONCLUSION: EGF is involved in the regulation of PS synthesis in adult rats under physiological conditions, and its signal transduction is related to the PTK receptor EGF and the intracellular PKC pathway
