采用生物热活性检测法,测定黏菌素作用于大肠杆菌的生物热活性谱,并以大肠杆菌生长时间延迟率Δt%为评价指标,建立黏菌素效价测定的新方法。结果显示,在17.0~41.6u·mL-1内,黏菌素浓度的对数值lgC与Δt%具有良好的线性关系(r=0.993);平均回收率为100.3%(n=9),且生物热活性检测法测定结果与管碟法测定结果一致(P>0.05)。结果表明,所建立的生物热活性检测法具有灵敏准确、实时在线、操作简便、干扰因素少、重现性好的特点,可作为黏菌素效价测定方法之一,同时所得生物热活性谱可弥补管碟法对细菌生长过程监控的不足。
The biothermic activity of colistin in E.coli was determined by bioactivity test, and a new method of determining the titer of colistin was established based on theΔt% of growth delay of E.coli. The results showed that in the range of 17.0 ~ 41.6u · mL-1, the logarithm of colistin concentration lgC had a good linear relationship with Δt% (r = 0.993); the average recovery was 100.3% (n = 9) The result of thermal activity test was consistent with that of plate and tube method (P> 0.05). The results showed that the bioassay method was sensitive, accurate, real-time on-line, easy to operate, less interference and reproducible. It could be used as one of the methods to determine the potency of colistin. At the same time, Can make up for the lack of monitoring the growth of bacterial growth plate method.
