目的:探讨瓜蒌皮提取物体外对高糖诱导人脐静脉内皮细胞(HUVECs)损伤的保护作用及机制。方法:体外培养人脐静脉内皮细胞株(HUVECs),采用MTT法观察12、24、48 h瓜蒌皮不同提取物对高糖损伤HUVECs细胞存活率的影响;收集24 h内皮细胞的上清液,检测细胞上清液中SOD、LDH活性及MDA含量。结果:与正常对照组比较,高糖损伤组细胞在24、48 h存活率显著降低(P<0.01),处理24 h后细胞上清液中SOD活性显著降低,LDH活性及MDA含量显著升高(P<0.01);与高糖损伤组比较,作用24 h瓜蒌皮各提取物组内皮细胞存活率显著提高,瓜蒌皮氯仿-甲醇提取物组细胞上清液SOD活性显著升高,LDH活性及MDA含量显著降低。结论:瓜蒌皮提取物对体外高糖诱导的HUVECs细胞损伤有保护作用,其中以氯仿-甲醇提取物作用最佳,其作用机制与抑制氧化应激有关。
Objective: To investigate the protective effect and mechanism of the extract from melon skin on the injury induced by high glucose in human umbilical vein endothelial cells (HUVECs) in vitro. Methods: Human umbilical vein endothelial cells (HUVECs) were cultured in vitro. MTT assay was used to observe the effects of different extracts of melon skin on the survival of HUVECs with high glucose at 12, 24 and 48 h. The supernatant of endothelial cells The activities of superoxide dismutase (SOD), lactate dehydrogenase (LDH) and MDA in supernatant were detected. Results: Compared with the normal control group, the viability of cells in high glucose group was significantly decreased at 24 and 48 h (P <0.01), and the activity of SOD and the content of MDA in the supernatant were significantly decreased (P <0.01). Compared with the high glucose group, the survival rate of endothelial cells in each group of extracts of Gualouguangpi for 24 h was significantly increased, and the activity of SOD in the supernatant of the chloroform-methanol extract group was significantly increased Activity and MDA content decreased significantly. Conclusion: The extract of melon skin protects HUVECs from injury induced by high glucose in vitro. Among them, chloroform-methanol extract has the best effect, and its mechanism is related to inhibition of oxidative stress.
