目的建立同时测定黄连-吴茱萸药对中盐酸小檗碱、吴茱萸碱和吴茱萸次碱的方法。方法采用HPLC法,色谱柱规格为Kromasil 100-5 C18(250mm×4.6mm,粒度5μm);流动相:甲醇-乙腈-十二烷基磺酸钠(SDS)5 mmol·L-1水溶液(2∶4∶4,p H 7.2);流速1 ml·min-1,柱温35℃,检测波长225 nm。结果盐酸小檗碱、吴茱萸碱和吴茱萸次碱的线性范围分别为0.4004~5.005、0.4324~5.405、0.2168~2.171μg,平均加样回收率分别为:99.01%(RSD2.22%)、99.37%(RSD1.94%)、98.68%(RSD1.64%)。结论此法操作简单,灵敏度高,可在225 nm处同时测定黄连-吴茱萸药对中3个有效成分的含量。
Objective To establish a method for the simultaneous determination of berberine hydrochloride, evodiamine and rutaecarpine in Coptis chinensis Franch. Methods The HPLC method was used. The column was Kromasil 100-5 C18 (250 mm × 4.6 mm, particle size 5 μm). The mobile phase consisted of methanol-acetonitrile-sodium dodecyl sulfate (SDS) 5 mmol·L- : 4: 4, p H 7.2); the flow rate was 1 ml · min-1, the column temperature was 35 ℃ and the detection wavelength was 225 nm. Results The linear ranges of berberine, evodiamine and rutaecarpine were 0.4004 ~ 5.005, 0.4324 ~ 5.405 and 0.2168 ~ 2.171μg, respectively. The average recoveries were 99.01% (RSD2.22%) and 99.37% RSD 1.94%), 98.68% (RSD 1.64%). Conclusion This method is simple and sensitive, and can be used to determine the content of three effective components of Coptis-Evodia rutaecarpa simultaneously at 225 nm.
