目的:观察麝香保心丸(SXBXW)对压力负荷性心室肥厚大鼠的作用。方法:用腹主动脉缩窄术制备压力负荷性心室肥厚模型,将实验大鼠随机分为手术组(含麝香保心丸亚组、美托洛尔亚组、心室肥厚亚组)、假手术组,手术组中前2亚组分别给予麝香保心丸13.5mg/kg、美托洛尔10mg/kg,心室肥厚亚组和假手术组给予等量0.9%氯化钠溶液灌胃,持续8周。8周后行心脏超声检查,处死并记录每只大鼠体重、全心重和左室重量(LVW),计算全心质量指数(HWI)、左心质量指数(LVWI)。用ELISA法检测大鼠血清脑钠肽(BNP)和肿瘤坏死因子-α(TNF-α)的含量;RT-PCR观察心肌组织BNP mRNA的表达水平。结果:麝香保心丸亚组大鼠的体重与其他各组相比,差异无统计学意义(P>0.05),但其全心重明显轻于心室肥厚亚组(P<0.05),而与美托洛尔亚组和假手术组相比,差异无统计学意义(P>0.05);其他重量指标如LVW、HWI、LVWI及心脏超声指标如室间隔收缩末厚度、室间隔舒张末厚度、左室后壁收缩末厚度、左室后壁舒张末厚度较假手术组和心室肥厚亚组均有改善(P<0.05),与美托洛尔亚组相比差异无统计学意义(P>0.05);血清BNP及心肌组织BNP mRNA的表达水平亦明显低于心室肥厚组(P<0.05),与假手术组和美托洛尔亚组相比差异无统计学意义(P>0.05)。各组血清TNF-α无明显差异。结论:麝香保心丸可以逆转压力负荷性心室肥厚大鼠的室壁厚度,改善心室重构,降低BNP的水平,对TNF-α的影响不明显,且与美托洛尔相比无明显差异。
Objective: To observe the effect of Shexiang Baoxin Pill (SXBXW) on pressure-overload ventricular hypertrophy in rats. Methods: The model of pressure-loaded ventricular hypertrophy was established by abdominal aortic constriction. The experimental rats were randomly divided into operation group (including musk-preserved heart pill subgroup, metoprolol subgroup and ventricular hypertrophy subgroup), sham operation In the first two subgroups of the operation group and the operation group, 13.5 mg / kg of Shexiang Baoxin Pill, 10 mg / kg of metoprolol, respectively, and 0.9% sodium chloride solution in the hypertrophic subgroup and the sham operation group were administered orally for 8 week. The expression of BNP mRNA in myocardium was detected by RT-PCR. Results: Compared with other groups, the body weight of Musk-Pill group was not significantly different (P> 0.05), but the total heart weight was significantly lower than that of the ventricular hypertrophy subgroup (P <0.05) There was no significant difference between the metoprolol subgroup and the sham operation group (P> 0.05). Other weight indexes such as LVW, HWI, LVWI and echocardiographic indexes such as ventricular end-systolic thickness, Left ventricular posterior wall end-systolic thickness, left ventricular posterior wall end-diastolic thickness than the sham operation group and ventricular hypertrophy sub-group were improved (P <0.05), compared with metoprolol subgroup there was no significant difference (P> 0.05). The levels of serum BNP and BNP mRNA in myocardium were also significantly lower than those in the ventricular hypertrophy group (P <0.05). There was no significant difference between the sham group and the metoprolol group (P> 0.05). There was no significant difference in serum TNF-α in each group. Conclusion: Shexiang Baoxin Pill can reverse the thickness of ventricular wall in pressure-overload ventricular hypertrophy rats, improve ventricular remodeling, reduce the level of BNP, and has no obvious effect on TNF-α, and there is no significant difference compared with metoprolol .
