目的:研究益气活血方药对肝星形细胞(HSC)骨骼系统的影响。探讨 HSC 骨骼系统与其功能的关系。方法:采用 ABC 免疫组化染色方法结合图像分析对猪血清诱导的不同造模时间的大鼠肝纤维化模型以及益气活血药物预防给药和治疗给药的肝星形细胞 Desmin、神经胶质酸性蛋白(GFAP)和α-平滑肌肌动蛋白(α-Actin)进行研究。结果:构成细胞骨架的主要成分 Desmin、GFAP 和α-Actin 在肝纤维化的发生中均发生变化,其中α-Actin 改变尤为明显。而益气活血药物预防组和治疗组α-SMA 阳性细胞数量明显低于5周纤维化模型组。结论:益气活血方药对 HSC 的骨骼系统可以产生影响。
Objective: To study the effect of Yiqi Huoxue Recipe on the skeletal system of hepatic stellate cells (HSC). Explore the relationship between the HSC skeletal system and its function. METHODS: ABC immunohistochemical staining and image analysis were used to determine the liver fibrosis model induced by pig serum at different time, and to prevent and treat the administration of hepatic stellate cells, Desmin and glial Acidic protein (GFAP) and α-actin were studied. Results: The main components of the cytoskeleton, Desmin, GFAP, and α-Actin, all changed during the development of hepatic fibrosis, and the change of α-Actin was particularly obvious. The number of α-SMA-positive cells in the prophylaxis group and the treatment group was significantly lower than that in the 5-week fibrosis model group. Conclusion: Yiqi Huoxue Recipe can affect the skeletal system of HSC.
