目的:观察麝香酮对易卒中型肾血管性高血压大鼠(RHRSP)血浆内皮素(endothelin,ET)和脑皮质微血管结构改变的影响。方法:复制肾血管性高血压大鼠动物模型,将雄性RHRSP自术后第8周分成两组:高血压组和麝香酮治疗组,将年龄、性别、数量相同的假手术组作对照组,测量收缩压(SBP)和12周时血浆ET含量,光镜和电镜下观察脑皮质微血管形态改变。结果:12周时麝香酮组的SBP水平比高血压组低[(23.95±0.73)kPa vs(26.13±2.22)kPa,P<0.05],ET水平也比高血压组低[(73.25±10.78)pg/mLvs(89.44±9.92)pg/mL,P<0.05]。光镜和电镜下观察均发现高血压组皮质微血管损害最严重,麝香酮组微血管形态损害比高血压组轻。结论:麝香酮可降低RHRSP的SBP和血浆ET水平,并能在一定程度上改善肾血管性高血压大鼠脑皮质微血管损害。
Objective: To observe the effect of muscone on plasma endothelin (ET) and cerebral microvascular structure in stroke-prone renovascular hypertensive rats (RHRSP). Methods: The animal model of renovascular hypertension was established. The male RHRSP was divided into two groups at the 8th week after operation: hypertension group and musk ketone treatment group. The sham operation group with the same age, gender and quantity as the control group, Systolic blood pressure (SBP) and plasma ET content were measured at 12 weeks. Morphological changes of cerebral cortex were observed under light microscope and electron microscope. Results: The SBP level in musk ketone group was lower than that in hypertension group at 12 weeks (23.95 ± 0.73 kPa vs 26.13 ± 2.22 kPa, P <0.05), and ET level was also lower than that in hypertension group (73.25 ± 10.78) pg / mL vs (89.44 ± 9.92) pg / mL, P <0.05]. Light microscopy and electron microscopy were found in the cortical microvascular damage in hypertensive group the most serious, muscone ketone microvascular damage than the hypertensive group lighter. Conclusion: Musk ketone can reduce the level of SBP and plasma ET in RHRSP and can ameliorate the microvascular damage of cerebral cortex in renovascular hypertensive rats.
