目的:充分利用冬瓜子自然资源,分离出高纯度植物甾醇,并最大限度地提高其提取率。方法:在单因素水平基础上,采用响应面的中心组合设计,考察分析液料比、皂化浓度、皂化时间及皂化温度4个因素总甾醇提取率的影响,对甾醇的提取工艺参数进行优化分析。结果确定最佳工艺参数为:液料比8.5:1(mL/g)、皂化浓度2.8 mol/L、皂化温度96℃、皂化时间65min,理论上最佳提取率为61.03 mg/100 g,实际提取率60.88 mg/100 g,相对误差为-0.25%,含量达96.9%。结论:该方法用于冬瓜子总甾醇的提取简便、稳定,可预测性强。
Objective: To make full use of winter melon seeds natural resources, isolated high purity phytosterols, and maximize its extraction rate. Methods: Based on the single factor level, the central composite design of response surface was used to investigate the effects of liquid-to-solid ratio, saponification concentration, saponification time and saponification temperature on the total sterol extraction rate, and the optimal extraction parameters of sterol . The results showed that the optimum technological parameters were as follows: the ratio of liquid to material was 8.5: 1 (mL / g), the saponification concentration was 2.8 mol / L, the saponification temperature was 96 ℃, the saponification time was 65min. The optimal extraction rate was 61.03 mg / The extraction rate was 60.88 mg / 100 g with the relative error of -0.25% and the content of 96.9%. Conclusion: The method is simple, stable and predictable for the extraction of total sterol from melon seeds.
