目的:观察补肾复方对压力负荷增加大鼠心肌α-MHC和β-MHC基因表达的影响,探讨补肾复方逆转心肌肥厚的作用机制。方法:将雄性SD大鼠随机分为假手术组、模型组、卡托普利组、补肾复方大剂量组、补肾复方小剂量组。造模4周后,再药物干预4周。采用RT-PCR的方法检测大鼠左室心肌组织中α-MHC、β-MHC基因表达。结果:模型组大鼠左室心肌组织α-MHC mRNA表达较假手术组下降(P<0.05),与模型组比较,卡托普利组、补肾复方大剂量组、小剂量组左室心肌组织α-MHC mRNA表达显著升高(P<0.05,P<0.01)。模型组大鼠左室心肌组织β-MHC mRNA表达较假手术组升高(P<0.05),与模型组比较,卡托普利组、补肾复方大剂量组、小剂量组左室心肌组织β-MHC mRNA表达下降(P<0.05)。结论:补肾复方可增加α-MHC基因表达,降低β-MHC基因表达,具有逆转心肌肥厚的作用。
Objective: To observe the effects of Bushen Recipe on myocardial α-MHC and β-MHC gene expression induced by pressure overload in rats and explore the mechanism of Bushen Recipe in reversing myocardial hypertrophy. Methods: Male Sprague-Dawley rats were randomly divided into sham-operation group, model group, captopril group, high-dose Bushen compound group and low-dose Bushen compound group. After 4 weeks of modeling, the drug intervention for 4 weeks. The mRNA expressions of α-MHC and β-MHC in left ventricular myocardium of rats were detected by RT-PCR. Results: Compared with the model group, the expression of α-MHC mRNA in left ventricular myocardium of model group decreased significantly (P <0.05). Compared with model group, the inhibitory effect of captopril, α-MHC mRNA expression was significantly increased (P <0.05, P <0.01). Compared with the model group, the expression of β-MHC mRNA in left ventricular myocardium in model group was significantly higher than that in sham-operated group (P <0.05). Compared with model group, -MHC mRNA expression decreased (P <0.05). Conclusion: Bushen Recipe can increase the expression of α-MHC gene and decrease the expression of β-MHC gene, which has the effect of reversing cardiac hypertrophy.
