为建立两株抗日本血吸虫单克隆抗—CA-7、CA-8,(均为IgM型)。用单抗DOT-ELISA方法可检测出血吸虫感染兔血清中的靶抗原。CA-7的靶抗原存在于虫卵、尾蚴中;CA-8的靶抗原存在于虫卵、尾蚴和成虫中。用亲和层析的方法分离纯化了这两株单抗的靶抗原。实验结果表明CA-7和CA-8的靶抗原是中性糖含量分别为23.3%和14.7%,Mr为220×103和200×103的糖蛋白。这两种抗原经高碘酸氧化后,都丧失与单抗结合的能力;但经蛋白酶消化后,仍保留抗原活性,表明与单抗CA-7、CA-8结合的位点均在糖蛋白的糖链上。CA-7靶抗原经碱水解去除O-糖链后,丧失与CA-7单抗结合的能力;CA-8靶抗原经碱水解后仍保留抗原活性,说明CA-7靶抗原决定簇位于O-糖链上,CA-8靶抗原决定簇位于N-糖链上。
To establish two anti-Schistosoma japonicum monoclonal anti-CA-7, CA-8, (both IgM type). The target antigen in sera of rabbits infected with Schistosoma japonicum can be detected by the monoclonal antibody DOT-ELISA method. The target antigen of CA-7 is present in eggs and cercariae; the target antigen of CA-8 is found in eggs, cercariae and adults. The target antigens of the two McAbs were isolated and purified by affinity chromatography. The experimental results showed that the target antigens of CA-7 and CA-8 were glycoproteins with neutral sugar contents of 23.3% and 14.7%, respectively, and Mr of 220 × 103 and 200 × 103. After these two antigens were oxidized by periodate, they both lost the ability of binding to the monoclonal antibody. However, the antigenic activity remained after protease digestion, indicating that the sites of binding to the monoclonal antibodies CA-7 and CA-8 were both in the glycoprotein On the sugar chain. The CA-7 target antigen lost its ability to bind to CA-7 after it was alkalized to remove O-glycan. CA-8 target antigen retains the antigenic activity after alkaline hydrolysis, indicating that CA-7 target epitope is located at O - On the sugar chain, the CA-8 target epitope is located on the N-sugar chain.
