通过体外实验,对Vc/FeSO4诱发的红细胞膜脂质过氧化(LP)发生条件及反应历程进行了探讨。结果表明:在37℃水浴45分钟内,红细胞膜蛋白浓度为0.75mg/ml,5.0μmol/LFeSO4,pH值7.4的5mmol/L磷酸盐缓冲液的条件下,较低浓度Vc(小于1.0mmol/L)时可促进红细胞膜脂质过氧化过程,但高浓度Vc(大于1.0mmol/L)则抑制红细胞膜脂质过氧化过程。Vc浓度为0.25mmol/L时,红细胞膜脂质过氧化产物丙二醛(MDA)于反应后45分钟内迅速上升,90分钟接近峰值,以后趋于平衡。揭示:一定比例的Vc/FeSO4可有效地激发红细胞膜LP。本研究初步建立了红细胞膜LP模型。
Vc / FeSO4-induced erythrocyte membrane lipid peroxidation (LP) conditions and reaction process were discussed in vitro. The results showed that under the condition of 37 ℃ water bath for 45 minutes, the membrane protein concentration of erythrocytes was 0.75mg / ml, 5.0μmol / L FeSO4, 5mmol / L phosphate buffer with pH 7.4, the lower concentration Vc ( Less than 1.0mmol / L) could promote lipid peroxidation of erythrocyte membrane, but high concentration of Vc (greater than 1.0mmol / L) inhibited lipid peroxidation of erythrocyte membrane. Vc concentration of 0.25mmol / L, erythrocyte membrane lipid peroxidation product malondialdehyde (MDA) increased rapidly within 45 minutes after the reaction, 90 minutes close to the peak, then tends to balance. Revealed: a certain percentage of Vc / FeSO4 can effectively stimulate the erythrocyte membrane LP. This study initially established the LP membrane model of erythrocytes.
