目的:通过检测痰热蕴肺型特发性肺纤维化大鼠应用清金益肺汤后血清超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)的活性及光镜下大鼠肺组织的病理变化,探讨清金益肺汤对于痰热蕴肺型特发性肺纤维化病程中氧化/抗氧化失衡的调节作用。方法:清洁级健康雄性SD大鼠40只,随机分为5组:空白对照组(K组)、痰热蕴肺型特发性肺纤维化模型组(M组)、中药治疗组(Z组)、激素治疗组(J组)、中药加激素治疗组(H组)。采用气管内滴博来霉素的方法建立肺纤维化模型,造模结束后第二天滴加脂多糖建立痰热蕴肺证模型。光镜下观察各组大鼠肺组织的病理改变;化学荧光法检测各组大鼠腹主动脉血清SOD、GSH的活性。结果:M组、Z组、J组、H组在光镜下均有肺纤维化的特征性病理改变,但程度有所不同。与K组相比,M、Z、J、H组SOD、GSH的活性明显降低,有显著统计学意义(P<0.01)。与M组相比,Z、J、H组SOD、GSH活性显著升高(P<0.01);且H组明显高于Z组和J组。J组SOD活性明显低于Z组(P<0.01),而J组和Z组的GSH活性相较没有统计学意义(P>0.05)。结论:痰热蕴肺型特发性肺纤维化大鼠肺内存在氧化应激,清金益肺汤可明显增加肺内SOD、GSH的含量,使其在抗氧化机制中发挥重要作用,从而说明清金益肺汤可纠正痰热蕴肺型肺纤维化的氧化/抗氧化失衡。
OBJECTIVE: To detect the activities of serum superoxide dismutase (SOD) and reduced glutathione (GSH) in rats with phlegm-damp lung-type idiopathic pulmonary fibrosis after application of Qingjin Yifei Tang, Pathological changes in rat lung tissue to explore the Qingjin Yifei Decoction on the phlegm type lung injury in idiopathic pulmonary fibrosis in the regulation of oxidative / antioxidant imbalance. Methods: Forty healthy male Sprague-Dawley rats were randomly divided into 5 groups: control group (K), model group with idiopathic pulmonary phlegm (M group), group Z with TCM ), Hormone treatment group (J group) and Chinese medicine plus hormone treatment group (H group). Pulmonary fibrosis model was established by intratracheal instillation of bleomycin and the model of phlegm-damp lung accumulation was established by adding lipopolysaccharide on the second day after modeling. The pathological changes of lung tissue in each group were observed under light microscope. The activity of SOD and GSH in serum of abdominal aorta of rats in each group were detected by chemical fluorescence method. Results: The characteristic pathological changes of pulmonary fibrosis were observed in M group, Z group, J group and H group under the light microscope, but the degrees were different. Compared with K group, the activities of SOD and GSH in M, Z, J and H groups were significantly decreased (P <0.01). Compared with M group, the activities of SOD and GSH in Z, J and H groups were significantly increased (P <0.01), and those in H group were significantly higher than those in Z and J groups. The SOD activity in J group was significantly lower than that in Z group (P <0.01), while the GSH activity in J group and Z group was not statistically significant (P> 0.05). CONCLUSION: Oxidative stress exists in the lungs of rats with phlegm-dampness-heat-induced idiopathic pulmonary fibrosis. Qingjin Yifei Decoction can significantly increase the contents of SOD and GSH in the lungs and play an important role in the anti-oxidative mechanism. Description Qingjin Yifei Decoction can correct the phlegm lung type pulmonary fibrosis oxidation / antioxidant imbalance.
