目的:探讨TNF抗大肠癌的作用及其机制。方法:建立人结肠腺癌裸小鼠移植瘤模型,观察rhTNF对人结肠腺癌SW-480裸小鼠移植瘤重量、组织学和超微结构及细胞周期动力学的影响。结果:rhT-NF对人结肠腺癌裸小鼠移植瘤具有抑制作用,抑瘤作用呈剂量依赖性;结肠癌细胞围绕血管出现片状坏死,并有大量炎细胞浸润;rhTNF能明显抑制结肠癌细胞从G0 /G1 期过渡到S和G2M 期,使细胞增殖指数显著降低,DNA抑制率呈剂量依赖性。结论:rhTNF对大肠癌生长具有抑制作用,可能与rhTNF诱导的炎症反应及血管损伤有关;rhTNF能特异性抑制大肠癌细胞从G0/G1 期向S和G2M 期过渡,进而抑制DNA合成
Objective: To investigate the effect of TNF on colorectal cancer and its mechanism. METHODS: A nude mouse xenograft model of human colon adenocarcinoma was established. The effects of rhTNF on the weight, histology, ultrastructure and cell cycle kinetics of human colon adenocarcinoma SW-480 nude mice were observed. RESULTS: rhT-NF had inhibitory effects on human colon adenocarcinoma in nude mice, tumor inhibition was dose-dependent, colon cancer cells showed necrosis around the blood vessels, and a large number of inflammatory cells infiltrated; rhTNF can significantly inhibit colon cancer The transition from the G0/G1 phase to the S and G2M phases resulted in a significant decrease in cell proliferation index and a dose-dependent inhibition of DNA. Conclusion: rhTNF has an inhibitory effect on the growth of colorectal cancer, which may be related to inflammation induced by rhTNF and vascular injury; rhTNF can specifically inhibit the transition of colorectal cancer cells from G0/G1 phase to S and G2M phase, thereby inhibiting DNA synthesis.
