目的检测脂联素mRNA在妊娠糖尿病患者脂肪及胎盘组织中的表达及在妊娠糖尿病发病机制中的作用。方法用半定量逆转录聚合酶链反应(RT-PCR)技术检测60名正常产妇及60例妊娠糖尿病(GDM)患者剖宫产后脂肪及胎盘中脂联素基因在转录水平的表达。结果 GDM患者脂肪组织中脂联素mRNA表达量为(1.05±0.12),正常妊娠者为(1.28±0.14),GDM患者低于正常妊娠组,差异有统计学意义(P<0.01);脂肪组织中脂联素mRNA表达量与GDM胰岛素抵抗指标相关(P<0.01);GDM患者与正常妊娠患者胎盘组织中脂联素mRNA表达差异无统计学意义(P>0.05)。结论脂肪组织脂联素表达的减少可能是GDM患者胰岛素抵抗的重要原因;胎盘脂联素的表达与GDM及其胰岛素抵抗关系不明显。
Objective To detect the expression of adiponectin mRNA in adipose tissue and placenta of gestational diabetes mellitus patients and its role in the pathogenesis of gestational diabetes mellitus. Methods The expression of adiponectin gene in adipose and placenta of cesarean section in 60 normal pregnant women and 60 cases of gestational diabetes mellitus (GDM) was detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). Results The adiponectin mRNA expression in adipose tissue was (1.05 ± 0.12) in GDM patients and (1.28 ± 0.14) in normal pregnancies, lower in GDM patients than in normal pregnancy group (P <0.01) The mRNA expression of adiponectin in GDM patients was positively correlated with GDM insulin resistance (P <0.01). There was no significant difference in adiponectin mRNA expression between placenta and GDM patients (P> 0.05). Conclusion The decrease of adiponectin expression in adipose tissue may be the important reason of insulin resistance in GDM patients. The expression of adiponectin in placenta is not related to GDM and insulin resistance.
