目的探讨淫羊藿苷对异丙肾上腺素诱导的小鼠肥大HL-1心肌细胞维生素D受体mRNA相对表达量的影响。方法以2×10~4·ml~(-1)密度接种细胞,用异丙肾上腺素诱导小鼠HL-1心肌细胞损伤,实验分为正常组、模型组、淫羊藿苷低剂量组、淫羊藿苷中剂量组、淫羊藿苷高剂量组、维生素D组。MTT法检测各组HL-1心肌细胞存活率。RealTime PCR法检测各组HL-1心肌细胞维生素D受体mRNA相对表达量。结果 MTT法显示不同剂量的淫羊藿苷均可提高小鼠肥大HL-1心肌细胞的存活率,且淫羊藿苷的浓度在10μmol·L-1时,细胞存活率最高。Real-Time PCR检测结果显示,与模型组比较,不同剂量的淫羊藿苷均可明显下调(P<0.05)异丙肾上腺素诱导的小鼠肥大HL-1心肌细胞维生素D受体mRNA相对表达量。结论淫羊藿苷对小鼠肥大HL-1心肌细胞具有保护作用,其机制可能通过维生素D轴调节VDR mRNA表达水平有关。
Objective To investigate the effect of icariin on the expression of vitamin D receptor mRNA in HL-1 cardiomyocytes induced by isoprenaline in mice. Methods Cells were inoculated at a density of 2 × 10 ~ 4 · ml ~ (-1) and induced by isoprenaline. The experiment was divided into normal group, model group, icariin low dose group, Icariin middle dose group, icariin high dose group, vitamin D group. MTT assay HL-1 cardiomyocyte survival rate of each group. RealTime PCR method was used to detect the relative expression of vitamin D receptor mRNA in HL-1 cardiomyocytes. Results MTT assay showed that different doses of icariin could increase the survival rate of HL-1 cardiomyocytes in hypertrophied mice, and the cell survival rate was the highest at 10μmol·L-1 icariin. Real-Time PCR results showed that Icariin at different doses significantly decreased (P <0.05) the relative expression of vitamin D receptor mRNA in HL-1 cardiomyocytes induced by isoprenaline in hypertrophied mice compared with model group the amount. Conclusion Icariin has a protective effect on hypertrophic HL-1 cardiomyocytes in mice, and its mechanism may be related to the regulation of VDR mRNA expression by vitamin D axis.
