目的建立人血液、尿液中保泰松的气相色谱氮磷检测的分析方法。方法在酸性条件下用乙酸乙酯提取血液、尿液中的保泰松,采用气相色谱法对其定性、定量分析。以正常人血液、尿液为空白样本,分别添加保泰松标准品,确定检材的前处理方法、色谱分析条件、工作曲线和线性范围,考察方法的精密度、回收率,并对1例实际案件的血液、尿液样本进行检测。结果该方法在10.0~800.0μg/m L范围内线性关系良好,血液和尿液的最低检测限分别为0.5μg/m L和1.0μg/m L(S/N≥3),日内精密度小于5%,日间精密度小于8%,方法回收率在98.6%~104.2%之间。结论本文方法便捷、准确度高,可以在血液、尿液中保泰松的检测中选用。
Objective To establish a method for the determination of phenylbutazone in human blood and urine by gas chromatography (RP-HPLC). Methods The geniposid was extracted from blood and urine with ethyl acetate under acidic conditions, and its content was determined qualitatively and quantitatively by gas chromatography. Taking normal blood and urine as blank samples, the standard of Phenylbutazone was respectively added to determine the pretreatment method, chromatographic conditions, working curve and linear range of the samples. The precision and recovery of the method were also investigated. One case The actual case of blood, urine samples for testing. Results The method showed a good linearity in the range of 10.0-800.0 μg / mL with minimal detection limits of 0.5 μg / mL and 1.0 μg / mL respectively (S / N≥3). The intra-day precision was less than 5%, day precision less than 8%, the recovery rate of 98.6% ~ 104.2%. Conclusion This method is convenient, accurate and can be used in the detection of phenylbutazone in blood and urine.
