目的:观察白藜芦醇(Res)对人前列腺癌细胞体外增殖的抑制作用,探讨Survivin和Bax在白藜芦醇抑制前列腺癌细胞体外增殖中的调节作用。方法:采用MTT法、流式细胞仪、免疫组化SP法观察了白藜芦醇对前列腺癌PC-3细胞增殖、细胞周期和蛋白表达的影响。结果:白藜芦醇对前列腺癌PC-3细胞增殖的抑制作用明显,呈浓度和时间依赖性。白藜芦醇使前列腺癌PC-3细胞周期阻滞于G0/G1期,且Bax蛋白表达上调,同时,Survivin蛋白表达下降,而Caspase-3活性升高。结论:白藜芦醇能抑制PC-3细胞的增殖,其机制可能是使前列腺癌PC-3细胞阻滞于G0/G1期,且Bax蛋白表达上升,同时Survivin蛋白表达下降,直接激活Caspase-3而抑制PC-3细胞的增殖。
OBJECTIVE: To observe the inhibitory effect of resveratrol on the proliferation of human prostate cancer cells in vitro and to explore the role of survivin and Bax in inhibiting the proliferation of prostate cancer cells in vitro. Methods: The effects of resveratrol on the proliferation, cell cycle and protein expression of prostate cancer PC-3 cells were observed by MTT assay, flow cytometry and immunohistochemistry. Results: Resveratrol significantly inhibited the proliferation of PC-3 cells in a concentration-and time-dependent manner. Resveratrol blocked the cell cycle of PC-3 cells in G0 / G1 phase and increased the expression of Bax protein. Meanwhile, the expression of Survivin protein decreased and the activity of Caspase-3 increased. CONCLUSION: Resveratrol can inhibit the proliferation of PC-3 cells. The mechanism may be that the PC-3 cells in the prostate cancer cells are arrested in the G0 / G1 phase and the expression of Bax protein is increased. Survivin protein expression is decreased, which directly activates the expression of Caspase- 3 and inhibit the proliferation of PC-3 cells.
