目的:观察肾衰泄浊颗粒对慢性肾功能衰竭(CRF)大鼠的影响并观察其急性毒性反应。方法:建立CRF大鼠模型,观察CRF大鼠使用肾衰泄浊颗粒前后肾功能及血液学指标的变化情况。将肾衰泄浊颗粒以最大浓度给小鼠灌胃,小鼠没有出现死亡,未测出LD50故测其最大耐受量。结果:肾衰泄浊颗粒能明显降低腺嘌呤所致CRF大鼠的血肌酐(Scr)、尿素氮(BUN)水平,能提高红细胞(RBC)、血红蛋白(HGB)和红细胞压积(HCT)。肾衰泄浊颗粒给小鼠灌服的最大耐受量可达46.16g/kg,相当于临床用量的276.96倍。结论:肾衰泄浊颗粒是治疗CRF大鼠的有效药物,且具有较好的安全性。
OBJECTIVE: To observe the effect of Shenshuai Xiezhu Granules on chronic renal failure (CRF) rats and observe its acute toxicity. METHODS: A rat model of CRF was established to observe the changes of renal function and hematological parameters before and after use of Renshao Xiezhuo granules in CRF rats. Kidney failure and turbid granules were given to the mice at the maximum concentration. The mice did not die. No LD50 was detected and the maximum tolerated dose was measured. RESULTS: Shenshuo Xiezhu Granules could significantly reduce serum creatinine (Scr) and urea nitrogen (BUN) levels in CRF rats induced by adenine, and increase red blood cells (RBC), hemoglobin (HGB), and hematocrit (HCT). The maximum tolerated dose of Renshuai Xiezhu Granules to mice can reach 46.16g/kg, which is equivalent to 276.96 times of clinical dosage. Conclusion: Shenshuai Xiezhu granules is an effective drug for treating CRF rats and has better safety.
