通过进一步分析采用全反式维甲酸(all trans retinoic acid, ATRA)诱导人肺腺癌 GLC-82细胞,经cDNA文库消减杂交构建的cDNA消减文库,得到一在ATRA诱导 GLC-82细胞分化过程中激活表达(retinoic acid induced, RAI)的基因的 cDNA片段,应 用菌落原位杂交技术在胎脑cDNA文库中进行筛选,克隆RAI的全长cDNA序列并测 序.RT-PCR分析表明,RAI在多种胎儿组织中表达.结果表明,RAI可能与细胞分化有 关,并在细胞基本生命活动中发挥重要作用.
By further analysis of all trans retinoic acid (ATRA) induced human lung adenocarcinoma GLC-82 cells by cDNA library subtractive hybridization cDNA subtractive library obtained a ATRA induced GLC-82 cell differentiation process Activation of expression (retinoic acid induced (RAI) cDNA fragments, using colony in situ hybridization in fetal brain cDNA library screening, cloning RAI full-length cDNA sequence and sequencing. RT-PCR analysis showed that RAI was expressed in a variety of fetal tissues. The results showed that RAI may be related to cell differentiation and play an important role in the basic life activities of cells.
