为研究水稻呼吸爆发氧化酶基因Os Rboh B在花药中特定的表达模式,构建了原位杂交探针载体PbskOs Rboh B,通过体外转录获得DIG标记的原位杂交探针,以不同发育时期的水稻花药为材料进行原位杂交试验。结果发现,Os Rboh B基因主要在水稻花药发育第8~10期的绒毡层和小孢子中表达,在第9期达到高峰,第10期开始下降。推测Os Rboh B主要参与水稻花药早期的发育,该研究为进一步研究Os Rboh B基因在水稻活性氧产生中的作用提供参考。
In order to study the specific expression pattern of Os Rboh B gene in rice anther, we constructed the in situ hybridization probe vector PbskOs Rboh B and obtained the DIG-labeled in situ hybridization probe by in vitro transcription. Anther materials for in situ hybridization test. The results showed that the Os Rboh B gene was mainly expressed in the tapetum and microspores from the 8th to 10th stage of anther development in rice. It reached its peak at the 9th stage and began to decline at the 10th stage. It is speculated that Os Rboh B is mainly involved in the early anther development of rice, which provides a reference for further study on the role of Os Rboh B gene in the production of reactive oxygen species in rice.
