从海南五指山原始森林、定安雷鸣发病胡椒园采集土壤样品及死亡线虫尸体,分离得到126株菌落形态有差异的真菌。以离体培养的南方根结线虫为靶标线虫对126株真菌进行筛选,获得7株对根结线虫校正死亡率在70%以上的菌株。菌株DF09002抗线虫活性最强,校正死亡率达100%;经5次继代培养,仍具有稳定产生杀线虫活性物质的能力。通过形态学和rDNA ITS序列分析,将该菌株归类为曲霉属黑色组曲霉黑曲霉集合体。酶系分析试验表明,菌株能产生在线虫侵染过程中起重要作用的几丁质酶和胞外蛋白酶,酶活力分别达到0.674、1.63 U/mL。
From the Wuzhishan virgin forest in Hainan province, the soil samples and dead coccidia were collected from the pepper farm in Ding’an County, and 126 strains of fungi with different colony morphology were isolated. A total of 126 strains of fungi were isolated from Meloidogyne incognita cultured in vitro, and 7 strains with a corrected mortality of more than 70% were obtained. The strain DF09002 was the most active against nematodes with a corrected mortality rate of 100%. After five subcultures, the strain DF09002 still had the ability to produce nematicidal actives stably. By morphological and rDNA ITS sequence analysis, the strain was classified as Aspergillus niger Aspergillus niger agglutinants. Enzymes analysis showed that the strains could produce chitinase and extracellular protease which play an important role in the process of nematode infection. The enzyme activity reached 0.674 and 1.63 U / mL, respectively.
