LOX对食管癌患者ESD术后吻合口狭窄处纤维细胞与瘢痕形成的影响

目的:分析沉默赖氨酰氧化酶(LOX)对食管癌患者内镜下黏膜剥离术(ESD)术后吻合口狭窄处纤维细胞与瘢痕形成的影响。方法:抽取洛阳东方医院2018年1月至2019年12月诊治的食管癌吻合口狭窄患者60例纳入观察组,另抽取同期无吻合口狭窄的食管癌患者60例纳入对照组。两组患者均接受ESD术治疗,行细胞迁移实验、实时荧光定量聚合酶链式反应(real-time PCR),评价瘢痕情况。结果:观察组SiRNA-LOX1、SiRNA-LOX2、SiRNA-LOX3表达高于对照组(n P<0.05)。观察组SiRNA-LOX3转染食管成纤维细胞后基质金属蛋白酶-2、基质金属蛋白酶-9的表达水平高于对照组(n P<0.05)。观察组患者LOX、纤维粘连蛋白mRNA表达水平高于对照组(n P<0.05)。观察组患者瘢痕评估量表评分高于对照组(n P<0.05)。n 结论:LOX对食管癌患者ESD术后吻合口狭窄处纤维细胞与瘢痕形成有促进作用,可稳定及增强细胞网状结构,增强细胞纤维能力,从而影响吻合口狭窄及瘢痕的形成。“,”Objective:To investigate the effects of Lysyl oxidase (LOX) on the formation of fibroblast and scar in anastomotic stenosis of patients with esophageal cancer after endoscopic mucosal dissection (ESD).Methods:A total of 60 esophageal cancer patients with anastomotic stenosis diagnosed and treated in Luoyang Dongfang Hospital from January 2018 to December 2019 were selected as the observation group, and another 60 esophageal cancer patients without anastomotic stenosis at the same period were selected as the control group. All patients received ESD. Cell migration assay and real-time polymerase chain reaction (PCR) were performed, and the scar was also evaluated.Results:The expressions of SiRNA-LOX1, SiRNA-LOX2 and SiRNA-LOX3 in the observation group was higher than those in the control group (n P<0.05). The expression levels of matrix metalloproteinase-2 and matrix metalloproteinase-9 in the observation group after transfection of esophageal fibroblasts with SiRNA-LOX3 were higher than those in the control group (n P<0.05). The expressions of LOX and fibronect in mRNA in the observation group were higher than those in the control group (n P<0.05). The observer scar assessment scale (OSAS) score of the observation group was higher than that of the control group (n P<0.05).n Conclusions:LOX can promote the formation of fibroblasts and scars at the anastomotic stenosis in patients with esophageal cancer after ESD. It can stabilize and strengthen the cell network structure and enhance the cell fibrosis, thus affecting the anastomotic stenosis and the formation of scars after surgery.
