目的本研究通过测定fibulin-5在维、汉族女性盆底器官脱垂(POP)患者阴道前壁结缔组织中的表达,探讨盆底支持组织结构和代谢的变化在POP发生、发展中的作用及维、汉POP患者fibulin-5蛋白表达有无差异性,进一步研究其有无种族差异。方法收集2013年7月-2015年10月新疆医科大学第一附属医院妇科收治的诊断为POP的72例患者为实验组,同期因良性疾病行子宫全切患者128例为对照组。应用免疫组化法和实时荧光定量(RT-PCR)法测定各组阴道前壁组织中fibulin-5蛋白水平的相对表达量。结果1.免疫组化法:Fibulin-5在实验组的表达量明显少于对照组,差异具有统计学意义(P<0.05);维、汉实验组之间比较,维、汉对照组之间比较差异均无统计学意义(P>0.05);2.实时荧光定量(RT-PCR)法:汉族实验组、对照组比较,维吾尔族实验组、对照组比较,维、汉实验组及对照组之间分别比较均有统计学意义(P<0.05)。结论1.Fibulin-5蛋白在维持盆底支持组织的功能上起重要作用。2.推测Fibulin-5蛋白表达存在着维、汉差异性,具体导致差异的原因仍需进一步研究探讨。
Objective To investigate the expression of fibulin-5 in the anterior vaginal connective tissue in patients with pelvic organ prolapse (POP) in female and Han nationality in China and to explore the role of changes in pelvic floor supporting tissue structure and metabolism in the occurrence and development of POP. Victoria and Han POP patients with fibulin-5 protein expression differences, further study of its racial differences. Methods Seventy-two patients with POP diagnosed in gynecology department of the First Affiliated Hospital of Xinjiang Medical University from July 2013 to October 2015 were selected as the experimental group, and 128 patients underwent hysterectomy due to benign diseases as the control group. The relative expression of fibulin-5 protein in the anterior vaginal tissue of each group was determined by immunohistochemistry and real-time fluorescence quantitative PCR (RT-PCR). Results 1. Immunohistochemistry: Fibulin-5 expression in the experimental group was significantly less than the control group, the difference was statistically significant (P <0.05); Victoria and Han experimental group comparison between the two groups (P> 0.05) .2. Real-time fluorescence quantitative PCR (RT-PCR): Han experimental group, control group, Uygur experimental group, control group, Victoria and Han experimental group and control group Respectively, were statistically significant (P <0.05). Fibulin-5 protein plays an important role in maintaining the function of pelvic support tissue. It is speculated that the expression of Fibulin-5 exists in Victoria and Han differences, the specific cause of the differences still need further study.
