目的:探讨复方蜥蜴散不同微粒组合剂(简称XY)创新模式(独特配方、不同微粒、糊剂给药)抗癌、阻癌及抑癌的分子生物学机制。方法:将120只SPF级SD大鼠随机抽取20只作为空白组,其余100只采用0.02mol/L的甲基硝基亚硝基胍(MNNG)溶液按5m L/kg灌胃等综合因素造模8周,成功制备胃癌前病变(PLGC)大鼠模型后随机分为模型组、复方蜥蜴散80目组、100目组、80目100目等量混合组(XY80组、XY100组、XY80’100组均为1.5g·kg-1·d-1)、维酶素组(0.3g·kg-1·d-1)共5组,每组13只,均作相应处理后应用免疫组化法检测其对胃黏膜组织P53和Survivin表达的影响。结果:各组大鼠胃黏膜组织P53和Survivin的平均吸光度(A):经统计学分析,模型组与空白组比较,差异有统计学意义(P<0.05);各治疗组与模型组比较,差异有统计学意义(P<0.05);XY各治疗组与维酶素组比较,差异有统计学意义(P<0.05),尤以XY80’100组A值最低,XY80组、XY100组与XY80’100组比较,差异有统计学意义(P<0.05);经Pearson相关分析,Survivin与P53表达呈正相关(P<0.05),相关系数r=0.899。结论:P53和Survivin在PLGC过程中具有协同作用;复方蜥蜴散不同微粒组合剂创新模式不仅配方独特,而且采用80目和100目等量混合和糊剂给药方式可进一步降低凋亡促进基因P53的突变,下调凋亡抑制基因Survivin的表达,从而实现有效干预PLGC和延缓癌变进程的作用。
OBJECTIVE: To investigate the molecular mechanism of anti-cancer, anti-cancer and anti-cancer in the innovative model (special formula, different particles and paste administration) of different compounds of Lizards Powder (XY). Methods: Totally 120 Sprague-Dawley (SD) SD rats were randomly divided into blank group (n = 20) and the other 100 rats were treated with 0.02mol / L MNNG solution The model rats were randomly divided into model group, compound lizard group with 80 mesh, 100 mesh, 80 mesh and 100 mesh equal mixing groups (XY80, XY100, XY80 ’ 100 in each group were 1.5g · kg-1 · d-1), and there were 5 groups in each group (0.3g · kg-1 · d-1). Thirteen rats in each group were treated with immunohistochemistry Method to detect the expression of P53 and Survivin in gastric mucosa. Results: The average absorbance of P53 and Survivin in gastric mucosa of rats in each group (A): By statistical analysis, the difference between model group and blank group was statistically significant (P <0.05); Compared with model group, (P <0.05). There was significant difference between XY treatment group and Vitexin group (P <0.05), especially in XY80’100 group, the lowest was in XY80 group, XY100 group and XY80 (P <0.05). According to Pearson correlation analysis, there was a positive correlation between the expression of Survivin and P53 (P <0.05), and the correlation coefficient r = 0.899. CONCLUSIONS: P53 and Survivin have synergistic effect in PLGC process. The innovative model of compound lizards with different microparticle combinations not only has a unique formula, but also uses the same amount of 80 mesh and 100 mesh mix and paste administration to further reduce the expression of apoptosis promoting gene P53 And downregulate the expression of Survivin, an inhibitor of apoptosis, in order to effectively intervene PLGC and delay the process of carcinogenesis.
